Giemsa staining produced heavy bands in the centromeric and pericentromeric regions of chromosome II and the X chromosome, as well as one band on the centromere of chromosome III, and a stained block outside the centromeric region.
The Giemsa stained blocks in the pericentromeric region of chromosomes II, III and X disappeared after C-banding, demonstrating that they are not formed from constitutive heterochromatin.
The Y chromosome, that appeared totally heterochromatic with Giemsa staining, presented an intensely stained band in the centromeric region after C-banding and was sub-metacentric (Figure 2).

Wright's stain is a technique in histology that is used to make the differences between cells visible under light microscopy.
Wright's stain is also used in cytogenetics to stain chromosomes on slides for visualization and diagnosis of sydromes and disease.
There are related stains known as the buffered Wright stain, the Wright-Giemsa stain, and the buffered Wright-Giemsa stain, and specific instructions depend on the solutions being used, which may include Eosin Y, Azure B, and Methylene Blue (some commercial preparations combine solutions to simplify staining).

Background: Modified Giemsa staining has been favoured by many researchers because it is easy to perform but, like many other stains, demonstration of the bacteria depends on its morphology.
In 2 cases immunostain could not demonstrate the bacteria but they were identified with modified Giemsa stain while in 5 cases the bacteria were identified by immunostain but not with modified Giemsa stain.
The positive predictive value of modified Giemsa stain was 93% CI 75 -98.8%) while the negative predictive value was 74% (CI 48.6 - 89.9).

Kept tightly stoppered and free of moisture, stock Giemsa stain is stable at room temperature indefinitely (stock stain improves with age).
Note: As alternates to this 45-60 minutes in 2.5% Giemsa stain, the smears could be stained for shorter times in more concentrated stains; one alternate is 10 minutes in 10% Giemsa; the shorter stains yield faster results, but use more stain and might be of less predictable quality.
To ensure that proper staining results have been achieved, a positive smear (malaria) should be included with each new batch of working Giemsa stain.

Stock solutions of Giemsa stain must always be diluted by mixing an appropriate amount of stain with distilled neutral or slightly alkaline water; buffered saline is preferred because it provides a cleaner background and better preservation of parasite morphology.
The staining solution must be freshly prepared by mixing 5 ml of stock solution with 100 ml of buffered water.

demonstrated the presence of mast cells by immunohistochemical staining with anti human tryptase antibody in gastric biopsies and concluded that density of mast cells was 2 to 3 times greater in H. pylori infected gastric mucosa than in negative normal stomach [4].
The specimens were assessed for infection and inflammation and stained with anti-human mast cell tryptase to count mucosal mast cells.
Although we did not performed immunohistochemical stains for mast cell detection, we propose that Giemsa stain is a reliable and easy staining method available in all laboratories and also has good interobserver correlation.
