Gotthard Kettler was of an old Westphalian family and the 9th child of the German knight Gotthard Kettler zu Melrich (mentioned 1527 1556) and his wife Sophie of Nesselrode.
Gotthard's older brother Wilhelm Kettler was bishop of Münster 1553 1557.
Gotthard Kettler married Anna, duchess of Mecklenburg (1533 4 July 1602) in 1566, the daughter of duke Albert the Fair of Mecklenburg and the princess Anna of Brandenburg.
