Guanylate cyclase is the intracellular enzyme responsible for the synthesis of cGMP from GTP (GTP pyrophosphate-lyase (cyclizing), EC 4.6.1.2).
Soluble guanylate cyclase is comprised of two subunits (a and b), with at least one heme prosthetic group loosely held in place with a histidine as the trans-axial ligand.
Soluble guanylate cyclase isolated from the human placenta was found to exist primarily in the less active 6-coordinate heme configuration, whereas the more active enzyme isolated from the bovine lung, was found to be in the 5-coordinate heme configuration.

Calcium concentration in the dark-adapted retinal rod outer segment is in the 200 to 600 nM range, and the guanylate cyclase of rod outer segments is thought to be activated in response to a fall in calcium concentration triggered by light.
We conclude from this study that rod outer segment guanylate cyclase, unlike any cyclase known so far, is capable of activation by two different types of calcium-binding proteins, one that activates in response to a decrease in calcium concentration, and the other, described here, which activates in response to an increase in calcium-concentration.

The biochemical characteristics of rat testicular guanylate cyclase were investigated and the activity and subcellular distribution of the enzyme was determined during testicular development.
Examination of the effects of metal ions, nucleotides, detergents and other in vitro activators on the activity of guanylate cyclase revealed that the testicular enzyme is similar in most respects to guanylate cyclase isolated from other mammalian tissues.

The invention relates to the administration of guanylate cyclase inhibitors, such as methylene blue, for the treatment of septic shock, cardiogenic shock, hypovolemic shock, obstructive shock, neuropathic shock and hypotension caused by excessive doses of nitrovasodilators.
The method of claim 1 wherein said guanylate cyclase inhibitor is selected from the group consisting of methylene blue, 6-anilino-5, 8-quinolinedione, N-methylhydroxylamine, hydroxylamine, ethacrynic acid and retinol.
Finally, methylene blue inactivates guanylate cyclase, the enzyme responsible for raising cyclic GMP levels, which is critical, as the cGMP second messenger is central to the hypotensive effect of NO.

A key enzyme in photoreceptor cells is the membrane- bound guanylate cyclase that catalyzes the synthesis of the intracellular transmitter cyclic GMP.
This activation pathway is localized in the photoreceptor-bipolar synapse and, therefore, does not interfere with the regulation of guanylate cyclase by GCAPs in the outer segment of the photoreceptor cell.
The guanylate cyclase in photoreceptor cells is the first example of a membrane-bound guanylate cyclase that responds to opposite Ca signals by providing two distinct binding sites for two different Ca -sensors.

Marletta's students have painstakingly picked apart the guanylate cyclase protein, in particular the exact binding site for NO. It turns out to be a heme molecule nearly identical to the heme that binds oxygen in hemoglobin to carry it through the blood stream to muscle.
In order to understand how guanylate cyclase is put together, Marletta and his students went in search of similar enzymes in the genomes of other organisms.
By knocking out several genes coding for parts of the guanylate cyclase enzyme, they were able to show that this enzyme was acting as an oxygen detector, primarily steering worms away from too much oxygen.
