  Logwood - LoveToKnow 1911
The colouring principle of logwood exists in the timber in the form of a glucoside, from which it is liberated as haematoxylin by fermentation.
By exposure to the air, especially in alkaline solutions, haematoxylin is rapidly oxidized into haematein, C16H120,, with the development of a fine purple colour.
This reaction of haematoxylin is exceedingly rapid and delicate, rendering that body a laboratory test for alkalis.

Haematoxylin and Counterstains © Woods and Ellis 2000
Haematoxylin is obtained from the logwood, Haematoxylon campechianum, a tree of the order Leguminosae (Genus Eucaesalpinieae) and so named because of the reddish colour of its heartwood (from the Greek haemato, blood, and xylo, wood) and young leaves.
Alum haematoxylin staining is also influenced by other factors, including the concentration and age of staining solutions as well as the fixation and processing to which the tissue was subject.
The haematoxylin, potassium or sodium alum and sodium iodate are dissolved in distilled water.

 RE: Hematoxylin
HAEMATOXYLIN AND HAEMATEIN Haematoxylin is a natural colouring matter obtained from the wood of a species of tree, the Haematoxylon campechianum, Linn which is indigenous to Mexico but is cultivated in the West Indies.
Confusion has sometimes arisen between haematoxylin and a crude resinous substance, known as hematoxylin, sometimes referred to as haematoxylin, which was at one time used in pharmacy and is still extensively used in the tanning industry.
Haematoxylin as used in biology is obtained by extracting the crude product, hematoxylin, with ether, while haematein, as already indicated is an oxidation product of haematoxylin.

The rationale of the technique is obscure; both iodine and ferric chloride with which the haematoxylin is combined, are oxidising agents and it is this property which probably accounts for the production of a fl dye with cationic properties rather than a simple dye- mordant-tissue mechanism.
Iron haematoxylin is used in preference to alum haematoxylin as it is more resistant to extraction by picric acid.

Blue Histology - more about Histological Stains
The most widely used staining technique is that of Haematoxylin and Eosin, commonly called H&E. In this method the nuclei of cells is stained by the haematoxylin whilst the cytoplasm is coloured by the eosin.
This chemical is readily oxidised to haematein and it is this oxidised form of haematoxylin, which is the colouring agent in the above technique.
Connective tissue and muscle are coloured differently because the staining affinity of the tissue may be affected by the dye molecular size, density of the tissue, pH of the dye bath and/ or the use of colourless "dyes".

Transfer the section to several changes of distilled water, a few minutes each, and then into a 0.5% w/v solution of periodic acid in distilled water where it is left for 5 minutes.
Check the Haematoxylin stain; if the tissue should be overstained or the surface of the resin has become stained with Haematoxylin, this may be corrected by a short differentiation in acid alcohol (0.5% HCl in 70% ethanol) and "reblueing" of the stained tissue.
Rinse the section in distilled water and counterstain it, using a 5% solution of Eosin Y in distilled water; leave the section in the Eosin stain for 30 minutes to 1 hour, wash briefly in running water and check the staining of the tissue.

Although most haematoxylin stains may be obtained ready for use from various manufacturers, many workers prefer to make up their own solutions.
