Haemolysis is the release of intracellular components from erythrocytes, thrombocytes and leukocytes into the extracellular fluid, i.e.
Haemolysis is visible as a red colouration of the plasma or serum after centrifugation of the sample.
Even if in vitro haemolysis occurs more frequent than in vivo, the latter is of greater clinical importance because of its pathological origin, and because the parameters influenced by haemolysis are relevant for diagnosis, follow-up and therapeutic monitoring of the diseases.

Haemolysis is more likely to occur in individuals with a hereditary deficiency of glucose-6-phosphate dehydrogenase, sickle cell anaemia and sickle cell trait.
Acute intravascular haemolysis is the most characteristic sign, particularly in persons with red cell glucose-6-phosphate dehydrogenase deficiency; it is accompanied by anaemia, leucocytosis, fever, haemoglobinuria, jaundice, renal insufficiency and sometimes disturbances in liver function.
Haemolysis mostly occurs in individuals with a hereditary deficiency of glucose-6-phosphate dehydrogenase due to instability of erythrocyte glutathion.

Haemolysis Artefact on blood Tests   (Site not responding. Last check: 2007-10-22)
Haemolysis, simply defined, is the process that leads to lysis (or rupture) of the RBC wall, causing the RBC to disintegrate and release its internal contents into the plasma, or fluid part of the blood.
The degree or severity of haemolysis is reflected in the intensity of the red colour of the plasma.
The effects of haemolysis on patients test results is somewhat test dependent and varies greatly with the degree of haemolysis in the specimen.

Haemolysis was detected in five of 137 procedures following implantation of Cook detachable duct coils.
Haemolysis is an important complication after duct coil implantation.
Haemolysis occurred in five children (two girls, three boys) with arterial ducts and otherwise normal hearts after coil implantation.

Haemolysis and Lipaemia   (Site not responding. Last check: 2007-10-22)
Haemolysis is a condition that occurs when haemoglobin is released from red blood cells when the cell membranes are damaged.
The fact that haemolysis has occurred may not be apparent until the separation of the serum or plasma has taken place.
Haemolysis will affect certain biochemical values when a colorimetric photometric method is used to determine these values.

Following substance-induced haemolysis, haemosiderin deposition can be accelerated to such levels that the accumulated pigment exceeds the normal amount of deposits in the spleen and may affect other organs as well.
During prolonged haemolysis, an increase in haemosiderin accumulation is expected to be seen in the bone marrow.

Haemolysis and haemoglobinuria are common observations in animals treated orally with EGBE for acute duration (ATSDR 1998).
Male rats receiving a single dose of 500 mg/kg by gavage exhibited haemolysis of erythrocytes accompanied by a drastic increase in the concentration of free haemoglobin in the plasma and haemoglobinuria was observed in all rats.
In male rats given 250 mg/kg by gavage, haemolysis was noted, characterised by an increase in mean cell volume and haematocrit, followed by a decline in haemoglobin concentration and RBC count.

The presence of haemolysis or lipaemia is indicated on the report and affected analytes are flagged.
Usually there is a false elevation in measured concentration, the exceptions include glucose (reduced due to haemolysis), calcium (reduced due to gross haemolysis) and creatinine (occasionally reduced due to haemolysis).

Haemolysis can be defined as damage to, or destruction of, red blood cells resulting in leakage of haemoglobin and red discolouration of the serum/plasma.
There are many causes of haemolysis, it can occur as a pathological event, as in the case of immune mediated haemolytic anaemias, however, the majority of haemolysis occurs either during or after blood collection.
As previously mentioned, haemolysis imparts a red colour to serum/plasma, the intensity of which is proportional to the degree of haemoglobin released, and this will interfere with photometric methods of biochemical analysis.

Comparison of p25 presequence peptide and melittin. Red blood cell haemolysis and band 3 aggregation.   (Site not responding. Last check: 2007-10-22)
Red blood cell haemolysis and band 3 aggregation.Aggregation cannot be reversed by high ionic strength; this serves to differentiate these peptides from other positively charged species such as polylysine that aggregate band 3 at low ionic strength.
The present study extends the results of leakage experiments on model-membrane systems to the haemolysis of human erythrocytes, which both peptides are shown to accomplish in a manner sensitive to membrane potential.
The 25 residue presequence (p25) for subunit IV of yeast cytochrome oxidase had previously been shown to possess structural and behavioural characteristics in common with the bee venom polypeptide, melittin.

The possibility that certain human subpopulations, including the aged and those predisposed to haemolytic disorders, might be at an increased risk from exposure to EGBE has been investigated using blood from the elderly, from patients with sickle cell disease, and from patients with spherocytosis.
These effects, which generally occur at higher exposure levels than those causing haemolysis, are secondary to the haematotoxicity of EGBE and a result of a compensatory response to the haemolysis.
Data indicate that humans are less sensitive to the haemolytic toxicity of EGBE than are rats as no or very slight haemolytic effects were observed in the poisoning cases after acute oral ingestion of large doses.

Results: In the first case, of a 75 year-old man with leukaemia and a retropharyngeal abscess, the isolate was consistently non-haemolytic, probably due to a failure to produce streptolysin S as has been described before in a small number of reports involving various M-types.
In the first patient there was a very narrow zone of greening around the colonies but no haemolysis in either aerobic or anaerobic cultures; failure to lyse red biood cells around the colonies in any part of the culture plate was confirmed by low power light microscopy.
However, zones of haemolysis were smaller where the colonies were more widely separated and, in the most thinly spread parts of the plate, the colonies showed small haloes of greening but no haemolysis (confirmed by microscopy), resembling those of the first patient.

British Journal of Biomedical Science: Bimodal role for divalent cations in the mechanism of EDTA cytolysis
The roles of both Ca 2+ and Mg+on haemolysis were studied further at a fixed EDTA concentration (10 mmol/L).
Calcium cations produced a dose-dependent increase in EDTA-induced haemolysis, up to a maximal level of 5 mmol/L. Thereafter, it produced a progressive reduction in haemolysis, in a dose-dependent manner, gradually approaching that of the control buffer level (Figure 1).
A practical application of this study would include the common observation of haemolysis, as shown by dark brown discolouration of plasma, when high levels of EDTA are used either as anticoagulant or enzyme inhibitor in blood samples.
