  Ayahuasca, Yage and Harmaline
Harmaline is about twice as toxic as harmine in most lab animals; the half-lethal dose (half the animals die) of harmine in dogs and mice is about 200 mg./kg.
When Naranjo gave harmaline and harmine in psychotherapeutic situations to city dwellers (people who had never been in the jungle), he observed that much of the imagery that was aroused had to do with snakes, panthers, jaguars and other large felines.
Harmaline and harmine are both crystalline, the first appearing as yellow and the second as green hydrochloride salts.

British Journal of Pharmacology - Modulation of voltage-gated channel currents by harmaline and harmane
Harmaline and harmane (Sigma, Taufkirchen, Germany) were freshly dissolved in the respective bath solution (Table 1) at a concentration of 1 mM.
Figure 1 illustrates the action of harmaline (a, c, e) and harmane (b, d, f) on voltage-activated calcium (a, b), sodium (c, d) and potassium channel currents (e, f) under control conditions and after a steady state of the effect had been reached.
While harmaline reduced the current by 25% (at lower voltages) to 45% (at more depolarised voltages) (Figure 4e), the effect of harmane increased from 50% at lower to 75% at higher depolarisations (Figure 4f).

Harmaline probes act as non-toxic vital stains or markers for intracellular pools of serotonin, and enable direct visualization of the cellular interactions through fluorescence microscopy and flow cytometry.
Since tricyclic antidepressants and other classes of psychoactive drugs function through affecting serotonin neurotransmitter systems interactions, the harmaline probe would be a highly valuable tool in developing and assessing the effectiveness of novel drugs.
Furthermore, since many disease states involve the serotonin system, the harmaline probe would also be useful in the research on the neuro-pharmacology of disease.

The separation of harmaline from harmine is based on the fact that when a warm aqueous solution of the hydrochlorides is alkalinized with ammonia, harmaline is liberated only after the decomposition of harmine hydrochloride is complete.
The appearance of harmaline is readly detected under the microscope since it consists of plates while harmine forms long needles.
The addition of ammonia, therefore, is stopped as soon as crystals of harmaline are detected, the harmine is filtered off and the harmaline recovered from the filtrate by the addition of ammonia, The bases are then further purified by recrystallization of their hydrochlorides.

Harmaline and harmine are both crystalline powders, the fromer being a yellow hydrochloride salt while the latter appears green.
This suggests that harmaline, which differs only in the position of the methoxy group, may derive it’s activity from the mimicry of a metabolite that is normally involved in the control of states of consciousness.
Studies have been done where harmaline and harmine were given in psychotherapeutic situations to city dwellers (those who have never been to the jungle).

In Europe and the Middle East, there are also plants which contain harmaline and DMT: Syrian Rue and the Giant River Reed.

Harmine and harmaline are hallucinogenic on their own with doses starting from around 300 mg (Naranjo 1967), but often cause physical side-effects such as nausea and tremors in this dose range.

In view of the high incidence of these symptoms, it is advisable to administer the drug when the patient has an empty stomach, and not to use more than 4 mg.

Harmine, harmaline, tetrahydroharmine (THH), and N,N-dimethyltryptamine (DMT) were quantitated in plasma from 15 healthy male volunteers after the ingestion of ayahuasca, a beverage that has been used for religious purposes in Brazil since pre-Columbian times.
A growing awareness of the interest in this ancient shamanistic practice in modern urban cultures and the widespread popular dissemination of the inebriant effects and type and sources of the plant admixtures used to prepare the beverage have provided additional impetus for this study.
Linearity was observed for harmine, harmaline, and THH within these respective ranges.

The purpose of the present study was to examine mechanisms of activity-dependent changes of cerebral blood flow (CBF) in rat cerebellar cortex by laser-Doppler flowmetry, using two synaptic inputs that excite different regions of the same target cell, and with different synaptic strength.
The apical part of Purkinje cells was activated by electrical stimulation of parallel fibers, while the cell soma and the proximal part of the dendritic tree was activated by climbing fibers using harmaline i.p.
The results suggest that CBF increases elicited by activation of Purkinje cells are partially mediated by the NO-cGMP system independent of the input function, but that adenosine contributes as well when climbing fibers are activated.
