Hybridomas were screened for reactivity with crude cryptococcal protein preparations by a bioassay which measured IL-2 production in the presence of antigen-presenting cells.
Following immunization of mice with live and glass bead- disrupted C. neoformans, 48 and 65 hybridomas were generated, of which 26 and 28, respectively, reacted with C. neoformans.
Thus, using T cell hybridoma technology, we have purified a protein from C. neoformans that stimulates a T cell response.

Hybridomas producing a monoclonal antibody of the IgG class are produced by cultivating a hybridoma producing a monoclonal antibody belonging to the IgM class in the presence of an antigen reactive with the IgM antibody, and in the presence of thymocytes.
This hybridoma production method makes it easy to obtain monoclonal antibodies of the IgG class to a glycolipid or the like to which monoclonal antibodies of the IgG class are difficult to obtain.
    This invention relates to a method of producing hybridomas which comprises converting (class-switching) a hybridoma capable of producing a monoclonal antibody belonging to the IgM class to a hybridoma capable of producing a monoclonal antibody belonging to the IgG class.

In addition to murine hybridomas, one may produce mixed species, or "heterohybridomas." Heterohybridomas are created by fusing splenocytes obtained from a species other than a mouse with a murine plasmacytoma.
Mice syngeneic to the hybridoma, or, alternatively, immunocompromised stocks or strains, are inoculated intraperitoneally with a priming agent, typically pristane, 7-21 days prior to intraperitoneal inoculation of hybridoma cells.
Our reasoning is: most hybridomas will grow and produce a suitable quality and quantity of antibody in vitro; one cannot predict whether a specific hybridoma will fail to produce antibody in vitro; technology is available to most laboratories for this purpose; and in vivo production may lead to pain or distress.

 HYBRIDOMA CORE - Services
Sub-Cloning and Cryopreservation:  Hybridomas assaying positive for desired antibody secretion are grown in successively larger wells and then subcloned in the presence of thymocyte feeder cells.  After growth of subclones the supernatants are rescreened.  Selected subclones are then expanded and cryopreserved in liquid nitrogen.
Human-Human Hybridomas:  These have been successfully produced in this facility using methods described by Rauch (8).  These methods, which use the human B lymphoblastoid line GM4672 as the fusion partner, as well as other methods, will be available on an ongoing basis in this core facility.
Rabbit Hybridomas:  The Hybridoma Facility will be acquiring the rabbit plastocytoma cell line 240E1.  Fusions of splenocytes from immunized rabbits with this cell line have successfully generated stable hybridomas secreting monoclonal rabbit antibodies (9).

The fusion partner used for murine hybridomas is the P3X63-Ag8.653 cell line developed by Kearney et al (3).
Hybridomas are plated in a highly enriched selection medium and macroscopic colonies appear within seven to 10 days.  In approximately two weeks the medium is acidified and ready for screening by the investigator’s laboratory.
This procedure may be introduced in the Hybridoma Core on an experimental basis in the future: please inquire regarding the status of the technology.
