Hypodontia is an anomaly that may result in dental malpositioning, periodontal damage, lack of development of maxillary and mandibular bone height and has significant psychological, aesthetic and functional consequences.
Hypodontia and oligodontia are classified as isolated or non-syndromic, where as hypodontia/oligodontia and syndromic hypodontia/oligodontia or hypodontia/oligodontia are associated with syndromes.
In familial hypodontia, the type of inheritance in the majority of families seems to be autosomal dominant with incomplete penetration and variable expressivity.

The aims of this study were to determine the frequency of hypodontia of permanent teeth in twins discordant or concordant for clefts in each type of cleft, to determine the concordance of hypodontia in mono- and dizygotic twins, and to compare the findings with some earlier data.
The prevalence of hypodontia was 37% for monozygotic and 32.7% for dizygotic twins, 16.7% in the noncleft and cleft lip groups, 41.1% in the cleft palate group, and 64.3% in the cleft lip and palate group.

Prevalence of hypodontia ranges from 10% to 68% in different cleft types in Finland, being 10% in cleft lip, 33% in cleft palate, 49% in unilateral, and 68% in bilateral cleft lip and palate groups, and even higher in twins with clefts (Ranta, 1986; Laatikainen et al, 1994).
A higher incidence of hypodontia in the maxilla has been reported, and has been suggested to be a result of the same factors as for the cleft (Ranta et al, 1988).
Hypodontia in the mandible is more frequent in Pierre Robin patients than in that of the cleft patients (Ranta, 1986).

The tooth-and-nail syndrome (Witkop's syndrome) is a rare autosomal dominant ectodermal dysplasia manifest by defects of the nail plates of the fingers and toes and hypodontia with normal hair and sweat gland function.
Children with conical teeth, hypodontia or anodontia without some other readily apparent etiology should undergo a detailed examination to determine if abnormal nail plates on the fingers and toes are present.

Researchers in the D'Souza laboratory at the UTHSC-Houston Dental Branch, in collaboration with researchers at Baylor College of Medicine, have identified a gene responsible for congenitally missing teeth (teeth that never developed) in a Houston family.
Hypodontia is characterized by the absence of six or fewer permanent teeth, while oligodontia is characterized by the absence of more than six permanent teeth.
The researchers' efforts revealed that a mutation in the PAX9 gene was responsible for a rare form of oligodontia, missing molars, in the Houston family (published in the January 2000 issue of Nature Genetics).

Introduction: The purpose of this retrospective study was to assess the prevalence and distribution of hypodontia in the permanent dentition, excluding the third molars, in a sample of Japanese orthodontic patients.

Objectives: To determine the prevalence and severity of hypodontia in the western Kentucky (WK) region in comparison to urban central Kentucky (UK), rural southeastern Kentucky Appalachia (SEKA) and previous USA studies.
The prevalence of hypodontia was WKO=10.86%, WKP=8%, UKO=13.33%, UKP=7.94%, SEKA orthodontic=11.11% and SEKA pediatric dentistry=10.16%.
Conclusions: This study demonstrated the average prevalence of hypodontia in Western Kentucky (9.4%) is similar to other parts of the state, and high compared to most US hypodontia prevalence studies.

We are continuing to identify additional families segregating specific classes of non-syndromic hypodontia of unknown etiology and will identify the gene(s) underlying hypodontia by genome-wide linkage analysis, candidate gene identification and mutation analysis.
Towards understanding the pathophysiology of various movement disorders, we are identifying the underlying genes, with the long-term goal of studying the regulation of these genes in the normal and disease state and to develop treatment regimens based on these findings.
