IL6 is a prototypic pleiotropic cytokine produced by a variety of cells, and acts on a wide range of tissues far beyond the immune system.
IL6 is a single chain protein of MW 21 to 28 kDa, depending on the cellular source.
Human IL6 cDNA predicts a precursor of 212 residues, whereas the secreted form is composed of 184 amino acids after cleavage of the single peptide.

 Abstract
The proposed role of interleukin 6 (IL6) in athero-thrombotic disease is to stimulate the acute phase reactants, fibrinogen and CRP, both themselves independent vascular risk factors, as well as having a proinflammatory role in the atherosclerotic plaque.
Our aim was to analyse the impact of IL6 promoter haplotype on IL6 expression in cell types relevant to atherosclerosis and in the atherosclerotic plaque itself.
Haplotype-specific IL6 expression was not determined by the -174G/C polymorphism alone, strongly suggesting that it would be optimal to genotype for the -597G/A, -572G/C and -373AnTn polymorphisms as well as -174G/C in future genetic epidemiological studies.

In the present study we demonstrate that the cultured human keratinocyte cell line (SK-v cells) harboring and expressing integrated human papillomavirus type 16 (HPV16) DNA sequences constitutively releases IL6, which is known as a pleiotropic immunoregulatory cytokine of potential antitumor properties.
The presence of IL6 activity in SK-v cell-conditioned media (SK-v CM) was demonstrated by tritiated thymidine incorporation into IL6-dependent B9 murine plasmacytoma cells.
IL6 did not affect SK-v cell growth; however, it significantly augmented NK cell activity of human peripheral blood lymphocytes against both K562 erytholeukemic and SK-v cells as assessed by 51Cr release assay.

METHODS: The IL6 promoter polymorphism was investigated in 81 Caucasian patients with surgically treated squamous cell vulvar cancer using pyrosequencing.
In a multivariate analysis, including tumor stage and lymph node involvement as established prognostic factors and the IL6 promoter polymorphism, lymph node involvement, and the presence of at least one mutant allele, but not tumor stage, were associated with increased disease-free and overall survival.
