The technique is called "immunophenotyping" for 2 reasons: 1) It is dependent on the activity of antibodies, which are immunological substances 2) It is used chiefly to identify lymphoid and hematopoietic cells, which are part of the immune system.
Immunophenotyping is used clinically chiefly to diagnose and categorize lymphomas and leukemias.
Immunophenotyping is helpful because in many instances different kinds of benign and malignant lymphoid cells resemble each other in routinely stained tissue sections and smears.

 Consensus Leukemia Immunophenotyping
The scope of flow cytometric immunophenotyping is to provide an objective and reproducible method for the diagnosis and therapy control of hematological malignancies, based on the immunological identification and characterization of clonally expanded abnormal precursor cells of the various hematopoietic lineages.
Immunophenotyping also contributes to the further characterization of AML subtypes, especially the M0 and M7 subtype, while the FAB-definition of M1 to M6 is still based mainly on morphology and cytochemistry.
The major goals of immunophenotyping in mature lymphoid neoplasias currently are, therefore, the assignment of abnormal cells to the B or T cell lineage, their maturational analysis, and the characterization of specific phenotypes which might be helpful for the subclassification of disease.

The fluorochrome-labeled cells are analyzed by using a flow cytometer, which categorizes individual cells according to size, granularity, fluorochrome, and intensity of fluorescence.
To ensure accurate results for specimens from HIV-infected persons, laboratories must validate their hematology instrument's ability to give the same result at time 0 and at the maximum time claimed by the manufacturer when using specimens from both persons infected with HIV and those not infected.

A total of 103 patients with lymphadenopathy and accessible lymph nodes seen by Ola Landgren, MD, and colleagues at the Karolinska Hospital and Institutet in Stockholm, Sweden, underwent both diagnostic procedures.
Immunophenotyping was performed on both FNAC and histopathological specimens.
