Indican - Wikipedia, the free encyclopedia
Indican is a colourless organic compound, soluble in water, naturally occurring in Indigofera plants.
Tryptophan is first converted to indole, then to indican by bacteria in the gut.
Indican is then excreted into the urine and from there into the diaper where, upon exposure to air, it is converted to indigo blue dye.

As shown in the figure, indican (1) (water-soluble), which is a glucoside contained in the indigo plants principally in their leaves, is hydrolyzed by.beta.-glucosidase, an enzyme which co-exists with the indican in the same plant, to form indoxyl (2) (water-soluble).
In case of the fresh indigo leaves, the indican (1) present in the leaves would not be hydrolyzed by the enzyme (.beta.-glucosidase) unless the leaves are beaten or smashed, while the juice of the fresh indigo leaves is susceptible to hydrolysis by the enzyme (.beta.-glucosidase).
The formulation composed primarily of the dye component (i) including the indican derived from indigo plants, or the formulation composed primarily of the.beta.-glucosidase enzyme component (ii) is preferably encapsulated in bags made of a multi-layered plastic film impermeable to water and oxygen or in glass containers, for the purposes of storage and transportation.

What is Indican?   (Site not responding. Last check: 2007-09-30)
The INDICAN made in the liver is transported to the kidneys for excretion.
Elevated INDICAN levels are generally considered to be the result of "dysbiosis," the technical term for overgrowth of unfriendly organisms in the gut.
Rather than relying on bacteria to oxidize the INDICAN, in the Obermeyer test urine is treated with ferric chloride and concentrated hydrochloric acid in a test tube, which oxidizes and joins two of the INDICAN molecules to form indigo blue.

 Indications Of Urine Going Wrong
In testing for indican, it is necessary to understand the significance of the different shades of color obtained.
In the type of red indicanuria the indican is not precipitated, as in the case of the blue forms, but the result is very pronounced reddish coloration of the urine in the tube above the surface of the chloroform (see figure), while the latter remains colorless.
Obermayer's test for indican, recommended by Green: Put in a test tube 1 cubic centimeter Obermayer's reagent (15 drops) and an equal part urine to be tested, add 2 cubic centimeters of chloroform and thoroughly shake.

Urine Indican Test (Obermeyer Test) Overview
Following absorption, indole is converted to 3-hydroxy indole (indoxyl or indican) in the liver, where it is then conjugated with potassium sulfate or glucoronic acid.
As most of the endogenous indoles have a side chain which prevents cleavage and are instead metabolized to skatole, the production of indicans (indoxyl potassium sulfate and indoxyl glucoronate) reflects bacterial activity in the small and large intestines.
Detection of indicans depends upon its decomposition to indoxyl and subsequent oxidation to indigo blue.

Indican (1H-Indol-3-yl-β-D-glucopyranoside) is thought a byproduct of plant metabolism; a similar "metabolic indican" is present in the urine of mammals.
Plant indican is an indole molecule with a sugar moiety (glucose) attached via a β bond.
During the height of the indigo industry, indican was extracted by immersing bundles of Indigofera in large shallow vats of water.

Indican is by-product of bacterial breakdown of dietary protein in the upper bowel.
Since the upper bowel does not normally house many bacteria, indican is present only at low levels in a healthy person.
An elevated level of urinary indican is an indication of upper bowel bacterial overgrowth or dysbiosis.

INDICAN is not itself carcinogenic, and its presence in the bladder does not increase the risk of bladder cancer.
However, INDICAN is among several uremic toxins that may stimulate the progression of certain kidney diseases.

We report here about the localization of the beta-glucosidase and indican in the cells on the basis of biochemical and electron microscopical analyses.
The content of indican in this organelle was found to be high enough to conclude that the localizations of beta-glucosidase and its substrate, indican, are different to each other.
Although we have no idea about the exact physiological significance of the separated existence of beta-glucosidase and indican in Indigo plant cells, it is not difficult to assume that they developed a system to avoid the suicide caused by insoluble indigo produced by the reaction between beta-glucosidase and indican, if they are not separated.
