Intein - Wikipedia, the free encyclopedia
An intein is a segment of a protein that is able to excise itself and rejoin the remaining portions (the exteins) with a peptide bond.
Inteins have been engineered for particular applications such as protein synthesis, and the selective labeling of protein segments, which is useful for NMR studies of large proteins.
The first part of an intein name is based on the scientific name of the organism in which it is found, and the second part is based on the name of the corresponding gene or extein.

Inteins have also been called "protein intron (intron: Sequence of a eukaryotic gene's DNA that is not translated into a protein) s".
Inteins are very efficient at protein splicing and they have accordingly found an important role in biotechnology (biotechnology: The branch of molecular biology that studies the use of microorganisms to perform specific industrial processes).
Inteins have been engineered for particular applications such as protein synthesis, and the selective labeling of protein segments, which is useful for NMR (NMR: Resonance of protons to radiation in a magnetic field) studies of large proteins.

Inteins are selfish genetic elements that excise themselves from the host protein during post translational processing, and religate the host protein with a peptide bond.
This intein is inserted in the same position as the inteins found in the ATPase A-subunits encoding gene in Pyrococcus abyssi, P. furiosus and P. horikoshii and is found 20 amino acids upstream of the intein in the homologous vma-1 gene in Saccharomyces cerevisiae.
Intein excision depends on the splicing domain of the intein and the first amino acid residue of the C-extein [8].

The intein protein-splicing C-terminal (C) domain is composed of the two adjacent motifs in the C-terminal 25-40 aa (including the conserved aa immediately C' to the intein).
Functional inteins with no EN domain (minimal inteins), the relation of the protein splicing domain to other Hint domains and the presence of different EN domains in inteins all indicate that the primeval inteins had no EN domains.
However, it is not clear in which cases this relation between the inteins is due to vertical transfer (the usual inheritance, from an organism to its progeny) or horizontal transfer (movement of DNA across species).

 Henry Paulus
Our aim is to elucidate the mechanism of catalysis of protein splicing by defining the catalytic center of the self-splicing intein, both in terms of the amino acid side chains involved and their arrangement in 3-dimensional space.
As a first step in our investigation, we cloned the RecA intein between two affinity tags as artificial exteins and genetically dissected away the portions of the intein that are involved in its homing endonuclease function so as to generate a minimal protein splicing element.
The excision of these inteins by protein splicing is required for the function of these proteins and the inhibition of protein splicing will therefore be lethal to this parasite.

The segment of the gene that encodes the intein is usually given the same name as the intein, but to avoid confusion, the name of the intein proper is usually capitalized, whereas the name of the corresponding gene segment is italicized.

After the recent discovery of an intein in Chilo iridescent virus [3], an insect-infecting NCLDV of Iridoviridae, this is the second report of an intein sequence in a eukaryote-infecting virus.
The splicing is defined by the self-catalytic excision of an intervening sequence ("intein") from a precursor host protein where it is located, and the concomitant ligation of the flanking amino- and carboxy-terminal fragments ("exteins") of the precursor.
A shift in the base compositions between intein and extein coding sequences is considered as indicating a recent acquisition of inteins [20].

Virology Journal | Abstract | A new example of viral intein in Mimivirus
Inteins are "protein introns" that remove themselves from their host proteins through an autocatalytic protein-splicing.
After their discovery, inteins have been quickly identified in all domains of life, but only once to date in the genome of a eukaryote-infecting virus.
The intriguing association of an extremophilic archaeal-type intein with a mesophilic eukaryotic-like PolB in Mimivirus is consistent with the hypothesis that DNA viruses might have been the central reservoir of inteins throughout the course of evolution.

Inteins are internal protein elements mediating post-translational protein splicing.
Inteins can artificially be split into an N-terminal half (Int-n) that contains the A and B motifs and a C-terminal half (Int-c) that contains the F and G motifs.
When the split intein fragments are fused with different protein or peptide precursors, they are able to conduct protein trans-splicing that assembles two separate precursor molecules into a mature hybrid protein.
