Interleukin-4 Suppresses IL-1–Induced Expression of Matrix Metalloproteinase-3 in Human Gingival Fibroblasts   (Site not responding. Last check: 2007-10-11)
Background: In periodontitis, matrix metalloproteinase-3 (MMP-3, stromelysin 1) is present at increased levels in active disease sites compared to inactive or healthy sites, and the levels are correlated with clinical parameters and associated with progression of the disease.
Interleukin (IL)-4 has been shown in human skin and synovial fibroblasts and articular chondrocytes to suppress IL-1–induced expression of MMP-3, but this has not been shown in human gingival fibroblasts.
The objective of this study is to determine the effects of IL-4 on the IL-1–induced expression of MMP-3 in human gingival fibroblasts isolated from patients with periodontitis.

Interleukin 4 (IL-4) is a potent mediator of growth and differentiation for various lymphoid and myeloid cells.
To isolate a cDNA encoding the murine IL-4 receptor, we have developed an expression cloning method that uses biotinylated ligand as a probe and that may be generally applicable to cloning of receptor genes.

Interleukin 4-targeted (IL-4-/-) mice are defective in T helper (Th)2 cytokine production as determined after nematode infection.
As Th2 cells appear to be selectively induced by oral immunization we investigated the ability of IL-4-/- mice to respond to perorally administered antigen.
Our findings demonstrate that IL-4 and probably functional Th2 cells are required for induction of gut mucosal antibody responses.

Description of ligand: Interleukin 4 (IL-4; AfCS ID A001262) is a pleiotropic type I cytokine expressed by Th2 T lymphocytes (T cells), basophils, and mast cells.
IL-4 may also be secreted by NK T cells, gamma/delta T cells, and eosinophils under some circumstances.
Like other type I cytokines, its tertiary structure is defined by a hydrophobic core consisting of 4 alpha helices.
