Iron-sulfur cluster - Wikipedia, the free encyclopedia
The 2 iron, 2 sulfur cluster, consisting of two iron atoms, with two inorganic sulfur atoms found between the irons and acting as bridging ligands.
The 3 iron, 4 sulfur cluster, which is a 4 iron, 4 sulfur cluster with one missing iron atom.
Iron-sulfur clusters can be involved in oxidation-reduction reactions, or can be involved in catalysis.

Mutational analysis of the mitochondrial Rieske iron-sulfur protein of Saccharomyces cerevisiae. III. Import, protease processing, and assembly into the cytochrome bc1 complex of iron-sulfur protein lacking the iron-sulfur cluster.
Insertion of the iron-sulfur cluster is not an obligatory prerequisite to processing of the protein to its final size.
Either the processing protease cannot distinguish between iron-sulfur protein with or without the iron-sulfur cluster, or insertion of the iron-sulfur cluster occurs after the protein is processed to its mature size, possibly after it is assembled in the cytochrome bc1 complex.
We have used site-directed mutagenesis of the Saccharomyces cerevisiae Rieske iron-sulfur protein gene (RIP 1) to convert cysteines 159, 164, 178, and 180 to serines, and to convert histidines 161 and 181 to arginines.

Iron-sulfur protein - Wikipedia, the free encyclopedia
Iron-sulfur proteins are proteins characterized by the presence of polymetallic systems (iron-sulfur clusters) containing sulfide ions, in which the iron ions have variable oxidation states.
The simplest polymetallic system, [Fe ] cluster, is constituted by two iron ions bridged by two sulfide ions and coordinated by four cysteinyl ligands (in Fe ferredoxins) or by two cysteines and two histidines (in Rieske proteins).
Another common polymetallic system, [Fe ] cluster, consists of four iron ions and four sulfide ions placed at the vertices of a cubane-type structure, coordinated by four cysteinyl ligands.

Inorganic iron is also found in the iron-sulfur clusters of many enzymes, such as nitrogenase (Synthesis of ammonia from nitrogen and hydrogen) and hydogenase.
Iron is a metal extracted from iron ore, and is hardly ever found in the free (elemental) state.
Iron is the most abundant metal, and is believed to be the tenth most abundant element, in the universe.

Iron sulfur cluster assembly in Saccharomyces cerevisiae involves the scaffold protein Isu, which is a substrate of the Hsp70 Ssq1 and the J-protein Jac1.
Therefore, it is proposed that in yeast the iron-sulfur cluster assembly pathway involves several methods of overcompensation.

 Dr. Ding - Homepage
Throughout evolution, iron-sulfur clusters have become integral parts of diverse cellular functions such as energy conversion, the citrate acid cycle, nitrogen fixation, intracellular iron homeostasis, heme and biotin biosynthesis, DNA synthesis and DNA repair, and regulation of gene expression.
Current research is to elucidate the redox reactions by which iron-sulfur clusters are modified by reactive oxygen species and the cellular repair mechanism for the modified iron-sulfur clusters.
Among the six proteins (IscS, IscU, IscA, HscB, HscA and Ferredoxin) encoded by the gene cluster iscSUA-hscBA-fdx, IscS is a cysteine desulfurase that catalyzes desulfurization of L-cysteine and provides sulfur for the iron-sulfur cluster assembly in IscU.

The identification of genes encoding putative Isc proteins in the genomes of all amitochondrial protists sequenced so far [15-19] suggests that this mitochondrial function might have been retained in all amitochondrial protists and may be a general functional feature of all mitochondrion-derived organelles [12,16,20-22].
The position of Rickettsia prowazekii IscS basal to the eukaryotes suggests that eukaryotic IscS proteins originated from the mitochondrial endosymbiont, since this bacterium is considered to be a close relative to the mitochondrial ancestor.

 ISU1 -
Mitochondrial functional interactions between frataxin and Isu1p, the iron-sulfur cluster scaffold protein, in Saccharomyces cerevisiae.
Depletion of the cysteine desulfurase Nfs1p, the ferredoxin Yah1p or the yeast frataxin homologue Yfh1p by regulated gene expression causes a strong decrease in the de novo synthesis of Fe/S clusters on Isu1p.
Hence Ssq1p/Jac1p and Grx5p are required in a step after Fe/S cluster synthesis on Isu1p, for instance in dissociation of preassembled Fe/S clusters from Isu1p and/or their insertion into apoproteins.

Binding of DnaK decreases the rates of forming IscU-Fd complexes (greater than 8-fold) and also causes a minor change on the rates of iron sulfur cluster transfer in IscU-Fd complexes.
The rate constants of cluster transfer from native ISA to Fd are shown to be similar to those of cluster transfer from Cys derivatives of ISA.
The factors that contribute to cluster transfer efficiency also appear to be similar, including the importance of cluster lability and solvent accessibility.
