An isocyanide, also called an isonitrile, is an organic compound with the functional group R-N≡C. The CN functionality is connected to the organic fragment via the nitrogen atom, not via carbon as is found in the isomeric nitriles, which have the connectivity R-C≡N. Hence the prefix iso.
Commonly, isocyanides are synthesized by the reaction of primary amines with dichlorocarbene or by dehydration of a formamide with phosphorus oxychloride.
Isocyanides are reactants in two multicomponent reactions: the Ugi reaction and the Passerini reaction.

The method comprises reacting a corresponding difunctional component (i.e., a.omega.-carboxyladehyde or a keto-acid), an amine and isocyanide in a nucleophilic polar protic solvent at a concentration suitable to form the lactam.
Isocyanides suitable for use in the method include those selected according to the substitutent, R, which is desired in the final product(s).

Previous work has shown, for short-chained isothiocyanates, a desulfurization (homolysis of the C-S bond) to form an isocyanide and triplet sulfur atom was the main mode of deactivation of the excited state.
Other studies have shown that for longer chains, such as benzyl isothiocyanate, that isomerization of the isothiocyanate to the thiocyanate form was the dominant reaction.
This is significant because isocyanide reagents are either expensive or not commercially available.

For the Passerini reactions the first position on the bar code was defined as the acid component, the second as the aldehyde and the third as the isocyanide.
