Yes, juke joint is a real expression, and the first element is the same as that in jukebox.
A juke joint, as it is usually spelled (though jook is also found), is 'a usually rural roadhouse providing liquor, music, food, prostitutes, etc.'.
These forms--juke, juke joint, and jukebox--were all first recorded in the 1930s; Zora Neale Hurston wrote about jukes in several of her books about fl folklore.

Juke Joint Jumpin' - An Aussie's Guide to Blues in the South
The juke joint, like most other aspects of genuine rural blues culture is in serious decline.
Tallying the precise number of existing juke joints is difficult for several reasons.
The point is that there could be any number of juke joints hidden in the swamps, woods and fields of the South and no-one but the local fl residents would know about it.

 Almind - History of Juke-Joints
The cafés were from the very beginning normally located next to the cotton fields and owned by the white first or second generation immigrated citizen and owner of the fields.
The expensive and often heavy mechanical musical instruments with coin-chutes were found in the better public places like oyster-bars etc. in the big cities, whereas the smaller and somewhat cheaper coin-op phonographs were distributed to amusement- and music-arcades, coffee houses, bars and saloons.
After the coin-op phonographs had been installed in the jukes or joints, that often were built as poor annexes to grocery stores in the rural areas, the term juke-box was brought into and accepted in the local language.

Imes focused his camera on nearly empty rooms, yet these bluesy, almost peopleless photographs capture fl cafes, roadhouses, and taverns as a fascinating folk art that resounds with energy and pulses with the joys and griefs of the clientele.
The names of these juke joints are almost as evocative as Imes's photographs--the Pink Pony in Darling, Mississippi, the People's Choice Café in Leland, Monkey's Place in Merigold, the Evening Star Lounge in Shaw, the Playboy Club in Louise, Juicy's Place in Marcella, the Social Inn in Gunnison, and A.D.'s Place in Glendora.

Despite the very temporary nature of these joints that the photos reveal I felt they showed something else though, an earthiness and honesty for the customers that is perhaps missing from the typical white bar with its respectable dcor and artificial facade.
I can just imagine the various bars in Juke Joint coming alive with the sound of raucous blues and relaxing good times, plate thirty-eight shows a hand-made sign in Juicy's Place, Marcella, saying 'BE NICE OR LEAVE'.
Juke Joint is well produced with a large photo per page and generous margins and my only criticism would be that there are not enough exterior photos of these amazing joints.
