  Karyology and tumorigenicity testing requirements: past, present and future.
At the present time, karyology and tumorigenicity are applied to primary, diploid, and continuous cell systems in an uneven and inappropriate manner, largely for historical reasons.
Karyology essentially has remained as a legacy of the intense debate that led to the acceptance of WI-38 cells.
Like karyology, tumorigenicity testing was incorporated into the assessment of diploid cells in an attempt to persuade regulatory authorities that the cells were normal and acceptable.

The advantage of karyology is the identification of fine differences in the karyotype (e.g., chromosome fragments, aneuploidy or trisomy).
FCM analysis on all groups will be accomplished three times: as embryos using karyology (next paragraph), and as larvae >250m (Chaiton and Allen, 1985) and newly settled spat (<3 mos old) using FCM.
The ability to assay individual larvae is a powerful tool for the analysis of initial proportions of mosaics because it gives us a window into ploidy composition of individuals before selective mortalities associated with metamorphosis.
