A clown today is one of various types of performer in the circus and rodeo, normally appearing in makeup, costume (typically large footwear, oversized clothing, bright colors and patterns), wig and fun nose, and enacting humorous sketches, usually in the interludes between major presentations.

Among the great characters the Fleischers animated in their hayday are Betty Boop and Popeye, but their first great character (I believe, at least) was Koko the Clown.
Basically Koko was little more than a spunky little guy the Fleischers used as a guinea pig to do experiments in wild animation, much like most of their other characters.
Besides the surreal landscape in the background and secondary animations constantly filling the screen, it's the flawless combination of psychedelic mutations and rotoscoped captures of Cab Calloway's actual dance motions (rotoscoping was a technique developed by the Fleischers to trace animation cels over live-action film) that make this scene so incredible.
