La Chapelle Sixtine prend son nom du Pape Sixte IV della Rovere (pape de 1471 à 1484) qui fit restaurer l'ancienne Cappella Magna entre 1477 et 1480.
La décoration des parois remonte au XVe siècle et comprend: les tentures en trompe l'œil, les Histoires de Moïse (parois sud - entrée) et du Christ (parois nord - entrée) et les portraits des Papes (parois nord - sud - entrée).
Vers la fin de l'année 1533 Clément VII de Médicis (pape de 1523 à 1534) demanda à Michel-Ange de modifier ultérieurement la décoration de la Chapelle Sixtine en peignant le Jugement Dernier sur le mur du fond de l'autel.
