Xianlin Han Abstract   (Site not responding. Last check: 2007-11-06)
Lipidomics is a rapidly expanding research field that uses analytical chemistry to investigate the biological function, significance, and sequelae of alterations in lipids and protein constituents mediating lipid metabolism, trafficking, or biological function in cells.
By employing ESI intrasource separation techniques (i.e., shotgun lipidomics) that we have developed, individual molecular species of most major and many minor lipid classes can be fingerprinted and quantitated directly from biological lipid extracts without the need for chromatographic purification.
The application of shotgun lipidomics to biomarker discovery in Alzheimer’s disease (AD) (i.e., the specific depletion of sulfatide mass content in brain tissues and cerebrospinal fluid at the earliest clinically recognizable stage of AD) will be presented.

Counting fats: new consortium to map cellular lipids
Brown is leading one of six national “lipidomics” centers, the consortium sites that will sift through and identify all of the lipids in the cell.
Phospholipids are the focus of Brown’s lipidomics center; they are especially important to cell signaling and account for a large portion of the cell’s lipid pool.
Another aspect of lipidomics that excites Brown is the idea of using “lipid profiles” as a way to further refine the molecular understanding of disease.

And therein lies the interest shown by the National Institutes of Health to award Obeid and Hannun—and MUSC—a Center of Biological Research Excellence (COBRE) grant, a five-year renewable affirmation to explore the roles of lipids in cell growth, cell death, cell aging, inflammation and infectious diseases.
The grant will allow the center to support the lipidomics research of junior faculty members as they are mentored by senior investigators and eventually "graduate" and themselves mentor new recruits to the program.
Also within the center will be three research cores: The lipidomics core to measure and synthesize lipids, an animal pathobiology core to develop specific animal models of disease, and a protein science core to allow people to work on the proteins that work on the lipids.

 Outlook: Lipidomics Research
In a matter of minutes, shotgun lipidomics can quantify and identify nearly all of the hundreds of lipids in a complex biological sample, even those present in minute amounts.
“ With shotgun lipidomics we can compare the lipid profile of various tissues during the healthy state to that of the disease state with amazing accuracy,” says Richard W. Gross, MD, PhD, the division’s director and a professor of medicine, chemistry and of molecular biology and pharmacology.
But the heart of shotgun lipidomics is a set of chemical techniques that ensure each lipid will appear on the instrument’s screen in a unique and predictable location, and this is where innovative thinking was needed.

At the Washington conference, Dr. Edward Dennis of the University of California-San Diego provided an overview of lipid researchers' major investigative resource—a collaboration of more than 30 researchers at 18 universities known as the Lipid Metabolites and Pathways Strategy (MAPS) Consortium, funded by the National Institute of General Medical Sciences.
"We plan to generate 'road maps' that will define how all the lipid components of a cell move through the complex lipidomic network, from biosynthesis to removal, including their important roles as secondary messengers," he explained.
Lipid MAPS is divided into focus areas that concentrate on major groupings of lipids, informatics, cell biology, lipid detection and quantitation, and lipid synthesis.
