Topic: Lipogenesis

In the News (Tue 29 Dec 09)

Mechanisms of nutritional and hormonal regulation of lipogenesis
Lipogenesis is stimulated by a high carbohydrate diet, whereas it is inhibited by polyunsaturated fatty acids and by fasting.
Lipogenesis encompasses the processes of fatty acid synthesis and subsequent triglyceride synthesis, and takes place in both liver and adipose tissue (Figure 1).
Lipogenesis entails a number of discrete steps, shown in the middle, which are controlled via allosteric interactions, by covalent modification and via changes in gene expression.

Obese women convert carbohydrate to fat faster than lean women
With the control diet, de novo lipogenesis was nearly twice the rate in the obese than in the lean subjects, regardless of the source of the carbohydrate.
Although rates of de novo lipogenesis in the carbohydrate-enriched energy-balanced control diet were significantly greater in the obese than in the lean women, the absolute quantities of fat synthesized from carbohydrate via de novo lipogenesis during both phases of the study were relatively small.

Myriceline — the patented herbal extract to prevent and remove cellulite.
Lipogenesis is a process that is aimed at storing energy, in the form of TG, into the adipocyte.
The later stimulates the translocation of transporter GLUT-4 from intracellular vesicles, where it is stored, to the cell membrane surface, thus increasing glucose intake (due to the lack of glycerol kinase into the adipocytes, glucose intake is essential for the synthesis of triglycerides).
Inhibiting lipogenesis (decreasing the rate of fatty acid and triglyceride synthesis, consequently reducing the formation of new fat).

Estimation of total lipogenesis and glycogenesis was conducted as previously described [4].
Over the 2 h postprandial period, there was approximately a 30% reduction in the amount of lipogenesis, exclusively in white adipose tissue, in response to a RS diet relative to a DS diet.
Decreased lipogenesis selectively in WAT and a reduction in adipocyte cell size in response to RS feeding have broad implications for the treatment and prevention of obesity.

 Increased Lipogenesis in Human Tumors
The present thesis forms an integral part of the efforts to understand the molecular mechanisms and the biological and clinical significance of increased lipogenesis in tumor cells.
In part I, it is explored whether the PI3-kinase/Akt pathway, a signal transduction cascade which is frequently dysregulated and constitutively activated in cancer cells, may contribute to increased lipogenesis.
In part II, the authors have tried to gain more insight into the functional significance of tumor-associated lipogenesis by unraveling the nature and fate of newly synthesized lipids in cancer cells.

Lipogenesis in mammals is inhibited by benzofurancarboxylic acids substituted on the aromatic ring by one of certain moieties, and the 2,3-dihydro counterparts thereof.
The data obtained from the tests are set out in Table 1,as the percent inhibition of lipogenesis compared to the results obtained in the control tests wherein only the test compound was omitted.
However, in general, satisfactory results are obtained when the acids are administered in a dosage of from about 1 to about 400 milligrams per kilogram of the animal's body weight.

Nutrition & Metabolism | Full text | Modulation of adipocyte lipogenesis by octanoate: involvement of reactive oxygen ...
Dietary medium-chain triglycerides (MCT) inhibit body fat mass growth in both animals and human [1-10] Early studies suggest that this effect might be caused by rapid absorption of MCT-derived MCFA and their β-oxidation in the liver, which reduces the circulating fatty acids available to the adipocytes [11].
This is correlated with inhibition of lipogenesis and reduction of lipogenic gene expression.
Compared to the pharmaceutical inhibitors of lipogenesis [68,69], the effects of octanoate can be considered as moderate and yet might be more desirable for physiological regulation of body fat mass without adversely affecting normal fat tissue functions.

On the other hand, suppression of 1,25-dihydroxyvitamin D with high calcium diets is thought to reduce intracellular calcium in fat cells, thereby decreasing lipogenesis and increasing lipolysis, resulting in an anti- obesity effect (19,22).
When these mice were fed a low calcium, high fat, high sucrose diet for six weeks, adipocyte lipogenesis increased, lipolysis decreased, and gain in body weight and adipose tissue accelerated (19).
In contrast, the high calcium diet markedly inhibited lipogenesis, increased lipolysis, and suppressed fat accretion and weight gain (19).

I LOUVEAU and F GONDRET: Hormonal regulation of lipogenesis in porcine adipocytes (Journal of Endocrinology)
The present study aimed to determine whether the regulation of lipogenesis by GH and/or insulin in porcine adipocytes also involved SREBP-1.
The increase was maximal at 1.7 and 17 nM in subcutaneous and perirenal adipocytes respectively, suggesting inter-depot differences in the regulation of lipogenesis by insulin.
Taken together, these data indicate that the regulation of lipogenesis by insulin and GH appears to not involve change in SREBP-1 mRNA level in porcine adipocytes.
