Analyses of the lipoxin analog's mechanism of action revealed a novel role for lipoxins in blocking the cellular cascade of dysfunctional immune responses that leads to inflammation in the mucosal lining of the gastrointestinal tract.
Lipoxin - technically called lipoxin A4 - is a lipid mediator (fatty acid derived molecule) generated by the immune system as part of the response to antigens and inflammation by lipoxygenase-dependent arachidonic acid metabolism.
Naturally occurring lipoxin A4 promotes the resolution of inflammation and modulates immune cell activity via potent agonist actions at lipoxin receptors on granulocytes, macrophages, dendritic cells, T lymphocytes, and epithelial cells.

HUM-MOLGEN: Reduced Lipoxin Linked To Damaged Cystic Fibrosis Lungs
Lipoxins are tiny lipids that 'switch off' the infection-fighting neutrophils that inadvertently damage the lung when releasing their toxic antimicrobials.
Lipoxins convert the immune response from one dominated by neutrophils to one dominated by lymphocytes and macrophages, whose attacks are more focused and produce less collateral damage.
When synthetic lipoxin was administered to these mice, they were able to decrease bacteria numbers faster and had reduced disease severity compared with control mice.

A "lipoxin analog" shall mean a compound which has an "active region" that functions like the active region of a "natural lipoxin", but which has a "metabolic transformation region" that differs from natural lipoxin.
Lipoxin analogs include compounds which are structurally similar to a natural lipoxin, compounds which share the same receptor recognition site, compounds which share the same or similar lipoxin metabolic transformation region as lipoxin, and compounds which are art-recognized as being analogs of lipoxin.

"The lipoxin analog is another step in our development program to broaden the therapeutic options for Crohn's patients." The laboratory findings appear in this week's edition of the Proceedings of the National Academy of Sciences.
Evolving research suggests that the primary defect in Crohn's disease may be a breakdown in the layers of mucosal barrier protecting the gastrointestinal tract.
In the research conducted by Berlex and its scientific collaborators on the colitis model of Crohn's disease, the company's oral lipoxin analog strongly attenuated mucosal markers of inflammation and also dramatically reduced the induction of IL-2, TNF-alpha and interferon gamma observed in the mucosa.

New studies demonstrate efficacy of novel Lipoxin analog in laboratory models of Crohn's disease   (Site not responding. Last check: 2007-11-02)
Berlex announced today that new laboratory results demonstrate that its modified version of the naturally occurring lipoxin molecule exerts potent anti-inflammatory and immunomodulatory effects in Crohn's disease in vivo models and may provide a promising new oral treatment.
Improvements in colitis symptoms -- weight loss, colon injury and mucosal inflammation -- for the oral lipoxin analog were similar to published findings for injectible anti-cytokine therapies and equal to oral prednisolone in a head-to-head pre-clinical comparison.
The lipoxin analog both prevented and treated colitis -- suggesting potential use in induction of remission and/or maintenance therapy in Crohn's disease.
