The Selenoenzyme Phospholipid Hydroperoxide Glutathione Peroxidase Controls the Activity of the 15-Lipoxygenase with ...
One general feature of the lipoxygenase reaction is the requirement for small amounts of hydroperoxy lipids acting as essential activators of the enzyme(19, 20).
The lipoxygenase activity with complex substrates was measured oxygraphically in a 1.6-ml oxygraphic chamber at 25 °C. Unless stated otherwise, the incubation mixture consisted of 0.1 M Tris-HCl buffer, pH 7.4, containing either submitochondrial particles (1 mg of protein/ml) or human LDL (0.5 mg of protein/ml) as substrate.
Lipoxygenases are known to require small amounts of hydroperoxy fatty acids as activators for their catalytic action(19, 20).

UCSC Biomedical Research Faculty - Professor Ted Holman
Lipoxygenases are enzymes implicated in a broad range of human cancers, as well as cardiac and inflammatory diseases.
These lipoxygenase products are the precursors of hormones, such as leukotrienes and lipoxins, which have been implicated as critical signaling molecules in a variety of inflammatory diseases and cancers.
Holman believes that the allosteric site may be critical to the regulation of lipoxygenase activity by either suppressing or activating the enzyme, and that the site might serve as a potential target for a novel class of lipoxygenase inhibitors.

E0051: Lipoxygenase - A Mysterious 'Purple' Form   (Site not responding. Last check: 2007-10-10)
Lipoxygenase belongs to a family of dioxygenases that catalyze oxygenation of polyunsaturated fatty acids by a redox reaction involving a non-heme iron cofactor, molecular oxygen, a diene system from a fatty acid and the enzyme's amino acids that support this process.
'Purple' lipoxygenase has been described[2] as an uncharacterized metastable state of enzyme triggered by addition of substrate and oxygen or excess product (13HPOD) to a solution of enzyme.
In this presentation we report recent results of x-ray analysis of the 'purple' form of soybean lipoxygenase-3[3] obtained by penetration of 13-HPOD into the crystals.
