MacConkeys agar (MAC), for example, supports preferentially the growth of Gram negative organisms and inhibits the growth of most Gram positive organisms due to the presence of crystal violet and bile salts.
Inoculate each pure culture and the mixed culture to a separate agar plate by flaming the inoculating loop in the bunsen burner and then transferring a loopful of bacterial inoculum from the broth culture to the plate.
Subculture one isolated colony to a second agar plate and streak for isolation as indicated above.

The purpose of my project was to determine what temperature grew bacteria the fastest whether it was 0, 25, or 37 degrees Celsius; it was also to see which bacteria grew on MacConkeys and Blood agar.
In conclusion to the growth on the different medias I found that E. coli and Pseudomonas aeruginosa were the only ones to grow on MacConkeys agar.
Since MacConkeys is a selective media and gram-negative bacteria only grow on it that means E. coli and Pseudomonas are gram-negative.

For genital cultures, we are wondering if it would be appropriate to bill for a Gardnerella screen (code 87081) along with code 87070.
We are currently using five plates for our genital cultures: Chocolate, MacConkeys, Blood, GC-Lect, and HBT-Bilayer to check for Gardnerella.
Recommendations regarding GC transport systems and have their been any comparisons between the JEMBEC and the Amies agar with charcoal transport swab in viability and recovery of the organisms within 24 to 48 hours.
