  Center for Medicago Genomics Research
Medicago truncatula (commonly called barrel medic or simply Medicago) is the primary model, or reference legume species for genomic and functional genomic research in the USA (Barker et al., 1990; Cook, 1999).
First, as a member of the Papilionoideae sub-family of the legumes, Medicago is closely related to the majority of crop and pasture legumes, its nearest cousin in this respect being alfalfa (Medicago sativa), the most important forage legume in the USA (Choi et al., 2004b).
An important sign that Medicago had come of age as a research model was the commencement of genome sequencing of this species.

Sequencing Medicago truncatula at the University of Oklahoma
Medicago truncatula is an annual relative of alfalfa (annual medic) but with a genome size of ~454 to 526 Mbp, is only half that of alfalfa.
The initial goal of the Medicago truncatula genome project is to generate approximately 1-fold whole genome shotgun sequence data from a double-stranded pUC-based genomic library and obtain target shotgun clones for additional primer walking-based sequencing after analysis vs the ~100,000 Medicago truncatula ESTs and the genome of Arabidopsis thaliana.

Since the phenomenon of plant gravitropism involves the coordinated response of several cells and tissues in a series of steps that include gravity sensing, signal transduction, and differential cellular growth, it is likely that the cytoskeleton is at least partially involved in each of the phases of gravitropism (Figure1).
Hairy roots of Medicago truncatula expressing GFP (A) and a red fluorescent protein-talin construct that binds to the actin cytoskeleton (B).
These efforts are greatly benefited by state of the art facilities for cellular imaging present at the Foundation that include a confocal microscope, a ratio fluorescence imaging system, fluorescent stereomicroscopes and a variety of microtomes for sectioning of fixed plant material.

Integrated structural, functional and comparative genomics of the model legume Medicago truncatula
The widely-used model plant Arabidopsis thaliana is unable to establish root endosymbioses and is thus not the most appropriate system for the study of this important aspect of plant biology.
Therefore, in the current project Medicago truncatula has been chosen as a model legume to study the symbiotic genetic programmes of plants and facilitate the genetics and breeding of important legume crops such as pea, faba bean, alfalfa and clover.
We propose a functional genomics project to identify most of the plant genes whose expression is modified in the course of symbiotic and pathogenic interactions, as well as other genes of agricultural importance which cannot be easily studied with A.
