Immediate Hypersensitivity to Methylparaben Causing False-Positive Results of Local Anesthetic Skin Testing or ...
Methylparaben was the only established cause for an immediate hypersensitivity reaction during LA ST identified in a large allergy practice during the past 16 years.
Methylparaben, one of the most commonly used parabens, is a well-documented cause of T-cell-mediated contact sensitivity.
Methylparaben is the preservative most commonly used in multidose vials, raising awareness that methylparaben is a potential cause for local reactions previously attributed to the anesthetic agent itself.

Development of a Multidose Formulation for a Humanized Monoclonal Antibody Using Experimental Design Techniques
The effect of antimicrobial parenteral preservatives (benzyl alcohol, chlorobutanol, methylparaben, propylparaben, phenol, and m-cresol) on protein stability was assessed using size-exclusion chromatography, differential scanning calorimetry, right-angle light scattering, UV spectroscopy, and potency testing using a cell-based fluorescence-activated cell sorting method.
The preservatives benzyl alcohol, chlorobutanol, methylparaben, and propylparaben were examined in the concentration ranges of 0 to 0.75%, 0 to 0.2%, 0 to 0.1%, and 0 to 0.01%, respectively.
The effects of 6 preservatives (benzyl alcohol, chlorobutanol, methylparaben, propylparaben, phenol, and m-cresol) on the stability of a humanized monoclonal antibody were examined in order to develop a multidose intravenous formulation.

Ageing claims put parabens back under the spotlight   (Site not responding. Last check: 2007-10-12)
Researchers in Japan say that methylparaben, a commonly used antiseptic agent for a range of cosmetics products, may cause skin to age when it is exposed to ultraviolet rays.
Because the product is included in daily applied powders, foundations, sun milk, and, ironically, anti-ageing products the researchers are particularly concerned by their findings.
The skin was then exposed to 30 millijoules of ultraviolet rays per square centimeter — an amount that is deemed to be about the average daily amount of exposure during summer weather.

Allergic reactions to local anesthetic agents may occur as a result of sensitivity to 1) either the ester or amide component, 2) the methylparaben used as a preservative in the multiple dose vials, or 3) the antioxidants used in some formulations.
Because of these possible hypersensitivity reactions, many manufacturers have reformulated some of their products to eliminate methylparaben.
A ruling by the Food and Drug Administration mandated the removal of methylparaben from all local anesthetic dental cartridges.
