Microscope slide - Wikipedia, the free encyclopedia
These were popular in Victorian England until the Royal Microscopical Society introduced the standardized micrtoscope slide in the form of a thin sheet of glass used to hold objects for examination under a microscope.
Since high power microscopes have a very narrow region within which they focus, the object to be viewed ("specimen") is typically placed on the middle of the slide with another, much thinner square (or circle or rectangle) of glass placed over the specimen.
For example, a graticule slide is a microscope slide that is marked with a grid of lines (for example, a 1 mm grid) that allow the size of objects seen under magnification to be easily estimated.

 Microscope Use
The microscope used is termed a bright-field microscope because the are observed or the microscopic field is brightly lighted and the objects being observed appear darker.
The resolving power of a microscope is a function of the wavelength of light used and the numerical aperture (NA), which is characteristic of the lens system and the medium between the lens and the specimen.
The safe procedure when using a strange microscope, and not knowing whether or not the microscope is parfocal, is to raise the body-tube by means of the coarse adjustment before attempting to shift from one power objective to another.

 How to use a microscope
Your microscope slide should be prepared with a coverslip or cover glass over the specimen.
Place the microscope slide on the stage and fasten it with the stage clips.
Move the microscope slide around so that the image is in the center of the field of view and readjust the mirror, illuminator or diaphragm for the clearest image.

Trials on scanning microscope slides of large subjects with a 35 mm Nikon Coolscan IV ED slide scanner.
The slot into which the 35 mm photo slide is inserted is a generous height so I made a simple modification to an empty plastic 35mm slide mount; the top half of the mount was removed and replaced with two cardboard pieces to guide and loosely hold the microscope slide in place.
As a novice at interpreting these sort of slides, the ability to view in a single wide field image the types, features and relative amounts of the component minerals was very useful, cf visual scanning of a much smaller field on a microscope slide.
I've started to scan some large subjects in my microscope slide collection just to enjoy viewing the mount in its entirety, in addition to visual scan viewing with the low power objective; it should be stressed that even the most modest 3.5x objective will resolve far more detail visually.

WebSlide - A Virtual Microscope Slide
Microscope slides can now be digitized in a way that makes common sense to view, with no compromise made for specimen sampling as the entire specimen can be scanned at high resolution with diagnostic quality.
Since the slide is scanned with low and high power objective magnifications, when you make a software objective change in our viewing software the magnification was done by optical means during the scan and is real, and the data reduction and scan speed induced by this technique is dramatic.
The typical procedure for scanning a microscope slide with the B L I S S Workstation is that the slide is initially scanned with a 1.25x microscope objective and a thumbnail image of the entire slide is built as shown below.
