State highways in Missouri were first designated in 1922 and Missouri's system was used (along with Wisconsin's) as a plan for the entire U.S. Highway System which was created in 1926.
Missouri has also changed highway designations with a US highway or an interstate with the same number is designated through the state (Missouri State Highway 40 was redesignated Missouri State Highway 14 to avoid duplicating numbers with U.S. Highway 40 which also passes through the state).
State highways in Missouri are the responsibility of the Missouri Department of Transportation.

U.S. 412 is duplexed with expressway-grade Cimarron Turnpike from Tulsa west to Interstate 35 and duplexed with the Cherokee Turnpike from 5 miles west of Choteau, Oklahoma to 8 miles west of the Arkansas state line.
As of 2004, the highway's eastern terminus is in Columbia, Tennessee at an intersection with Interstate 65.
US Highways 400 and 425 are similar violations of the standard numbering scheme.

U.S. Highway 54 in Miller County, Missouri, is and at all times relevant to this case was a federal-aid primary highway, approved as such by the Federal Highway Administration (or its predecessor, the Bureau of Public Roads) and by the Commission, and is part of the state highway system of Missouri.
The purpose of the 1965 Act was to regulate the use of billboard advertising adjacent to certain highways in the State, and to comply with the requirements of the Beautification Act.
The state highway commission shall give thirty days' notice by certified mail to the owner of the sign, display, or device which is illegal, to remove same if illegal, or to cause it to conform to the provisions of sections 226.500 to 226.600 if it is an authorized sign, display or device.

In 2004, a total of 28 people were killed and 1,167 were injured in traffic crashes involving construction or other work zones.
Missouri law requires drivers to slow their vehicle down and to move over when safe to do so when approaching emergency vehicles working on the highways.

A Colorado man has filed a wrongful death lawsuit nearly three years to the day after his son was shot and killed by a Missouri State Highway Patrol trooper.
Lester Wilson, citing mental pain and anguish suffered by his son, as well as funeral and burial expenses and the loss of his son's companionship, is seeking damages "in excess of $25,000" and other relief as the court deems appropriate.
According to the Highway Patrol's report, the incident began when a female motorist stopped at a gas station in Higginsville.
