Missouri State Highway 96 is a state highway running between Interstate 44 at Halltown and Missouri State Highway 171 near Carl Junction.
A large section of the highway between Halltown and Carthage served as U.S. Route 66 and is currently marked as Historic Route 66.
Missouri 96 was turned into Missouri Supplemental Route YY west of Missouri 171 when Kansas decommissioned the eastern part of Kansas 96.

In 1822, the state made each county responsible for maintenance of its roads and all free males between the ages of 16 and 45 were required to help.
In 1907, the state saw the need to appoint a State Highway Engineer under the Board of Agriculture to help counties with construction of roads that would someday be part of a state system.
