NMR spectroscopy - Wikipedia, the free encyclopedia
Much like using infrared spectroscopy to identify functional groups, analysis of a 1D NMR spectrum tells the scientist what atom environments (like a methyl proton), and in some cases how many atoms of each type, exist within the sample.
NMR is based in quantum mechanical properties of nuclei, and as such is very reliable, predictable and reproducible.
NMR spectroscopy is often the only way to obtain high resolution information on partially or wholly intrinsically unstructured proteins.

Nuclear magnetic resonance - Wikipedia, the free encyclopedia
NMR studies a magnetic nucleus, like that of a hydrogen atom, protium being the most receptive isotope at natural abundance, by aligning it with a very powerful external magnetic field and perturbing this alignment using an electromagnetic field.
NMR spectroscopy is one of the principal techniques used to obtain physical, chemical, electronic and structural information about a molecule.
Two important concepts for high-resolution solid-state NMR spectroscopy are the limitation of possible molecular orientation by sample orientation, and the reduction of anisotropic nuclear magnetic interactions by sample spinning.

NMR spectroscopy is based on transitions between nuclear spin states.
Modern NMR spectrometers with rf's of 100 MHz or greater employ fast Fourier analysis to transform the data from a time domain interferogram to a frequency domain spectrum.
In an NMR spectrum, the position of absorption by a proton depends on the net strength of the magnetic field that surrounds the proton.

 CHP - NMR - Introduction
Nuclear magnetic resonance (NMR) spectroscopy uses radiofrequency radiation to induce transitions between different nuclear spin states of samples in a magnetic field.
NMR spectroscopy can be used for quantitative measurements, but it is most useful for determining the structure of molecules (along with IR spectroscopy and mass spectrometry).
The utility of NMR spectroscopy for structural characterization arises because different atoms in a molecule experience slightly different magnetic fields and therefore transitions at slightly different resonance frequencies in an NMR spectrum.

It is aimed at people who are familiar with the use of routine NMR for structure determination and who wish to deepen their understanding of just exactly how NMR experiments work.
It demonstrates that in NMR it is possible, quite literally on the back of an envelope, to make exact predictions of the outcome of quite sophisticated experiments.
The experiments chosen are likely to be encountered in the routine NMR of small to medium-sized molecules, but are also applicable to the study of large biomolecules, such as proteins and nucleic acids.

 Two Dimensional NMR Spectroscopy
The invention of multidimensional spectra was the major leap in NMR spectroscopy apart from the introduction of FT-NMR.
C is very low and their gyromagnetic ratio is markedly lower than that of protons (see table).
Therefore, two strategies are used for increasing the low sensitivity of these nuclei: Isotopic enrichment of these nuclei in proteins and enhancement of the signal to noise ratio by the use of inverse NMR experiments in which the magnetization is tranferred from protons to the hetero nucleus.
