PAG-VIII: MOLECULAR GENETIC ANALYSIS OF THE HIGH OLEATE TRAIT IN PEANUT   (Site not responding. Last check: 2007-10-14)
This high oleate trait is recessive, and it exhibits one- or two- gene-segregation depending on the normal oleate varieties in crosses.
The objective of this study was to understand the molecular nature of the high oleate trait.
We examined the potential role of these two genes in the high oleate phenotype by analyzing peanut seeds for transcripts of each, comparing their nucleotide sequences in high and normal oleate varieties, and testing the enzyme activity of each in yeast.

Use of 3-aminotriazole to isolate oleate/acetate non-utilizing mutants of Aspergillus nidulans
Oleate non-utilizing (olu) mutants of Aspergillus nidulans were isolated on selective medium containing the catalase inhibitor, 3-aminotriazole (3-AT); in an attempt to obtain strains that would enable analysis of peroxisome biogenesis and function in filamentous fungi.
The selection procedure is based on the lethality of H2O2 formed during peroxisomal -oxidation of fatty acids in the presence of the catalase inhibitor 3-aminotriazole (3-AT).
The olu mutants were unable to grow on acetate as sole carbon source, suggesting that their lesions did not occur in the -oxidation pathway.

When yeast cells are transferred from a glucose containing medium to a medium containing the fatty acid oleate, expression of genes encoding peroxisomal proteins is highly induced.
We apply a novel technique, SAGE (1), for the qualitative and quantitative analysis of gene expression to identify genes that are highly expressed on oleate medium and regulated by Pip2p.
First results indicate that in wildtype cells grown on oleate the highest expressed gene (7%) is the ADH2 gene encoding cytoplasmic alcohol dehydrogenase.

Duodenal administration of casein and oleate increased plasma levels from oral administration of a poorly-water-soluble antiepileptic drug as compared to duodenal glucose and saline in a canine model.
Since duodenal oleate produced the most dramatic elevations in drug plasma levels as compared to noncaloric input, the effects of intravenous MK-329 prior to oral phenytoin and duodenal oleate were evaluated in a single dog study.
Inhibition of duodenal oleate's enhancement of phenytoin plasma levels by MK-329 pre-treatment in a single dog study is consistent with complete CCK antagonist inhibition of gallbladder contraction from duodenal (4) and oral (15) meal administration in man and duodenal lipid emulsion administration in dogs (17).

Additionally, there is concern that Oleate could be applied to sun- dried Natural raisins, and that the raisins could be represented as Oleates to circumvent the volume regulations that are typically in effect for Naturals.
First, the commenter contends that the Oleate varietal type was first created in 1981 through informal rulemaking, and that the rationale for creating the Oleate varietal [[Page 6917]] type in 1981 is the same as today for maintaining the varietal type (46 FR 39120; July 31, 1981).
In 1981, Oleates were considered relatively new to the U.S. industry and were developed to reduce the time required to sun-dry raisins and reduce problems associated with untimely rains.

We report here measurements of the surface hydrophobicity of commercial offset printing inks, and estimate the effects of sodium oleate (a dispersant), calcium oleate (a collector), and a nonionic surfactant.
For various minerals, the pH value of maximum flotation response (with oleate collector) is known to correlate with the minimum in the aqueous surface tension and a similar correlation may be valid for printing inks.
Microphotographs show that the shape of collected ink aggregates can be changed from sphere-like to fiber-like by adjusting the collector concentration, a surprising result which might be useful for improving the kinetics of flotation.
