  Metabolic poisons
Oligomycin, an antibiotic, acts by binding ATP synthase in such a way as to block the proton channel.
That is the mechanism by which oligomycin inhibits oxidative phosphorylation.
Experimentally, oligomycin has no effect on state IV respiration, that is, it has no direct effect on electron transport or the chemiosmotic gradient.

Examination of the three-dimensional structure of oligomycin B showed that it has a protruding structure attached to a circular structure (43) and the protruding portion, the length and width of which are around 7Å and 2Å, respectively, appears favorable for interacting with the Na occlusion site of Na/K-ATPase embedded in the transmembrane segments.

Abstract: The influence of perfluidone (37924133) on oligomycin inhibited mitochondrial respiration was investigated in-vitro.
Aliquots of 50 nanomolar adenosine-diphosphate, oligomycin, and perfluidone at concentrations from 0 to 160 millimolar (mM), or carbonylcyanide-p-trifluoromethoxyphenyl-hydrazone (FCCP), a classic protonophore, were added at specific time intervals.

Oligomycin also inhibited cell death induced by p53, in a cell line where p53 is known to induce transcription of the endogenous bax gene and to trigger apoptosis through a bax-dependent mechanism.
Moreover, oligomycin reportedly prevents the rise in membrane potential and matrix pH induced by mitochondria-dependent cell death stimuli such as Bax or staurosporine.

This process is catalyzed by an enzyme complex named ATP synthase, consisting of a catalytic segment F1, a proton-translocating segment Fo, and an intervening segment, stalk, that brings about the coupling of F1 and Fo segments.
As its name implies, OSCP renders the F1 enzyme sensitive to the antibiotic oligomycin, which results in the inhibition of the activity of F1.
It is hoped that this kind of analysis will lead to a better understanding of the mechanism of ATP synthesis in normal and diseased tissue, which may eventually help in diagnosing and possibly curing neurodegenerative diseases that involve defects in mitochondria.
