 Equal Temperament Tuning
Tuning the 1st and 6th strings: The E, open 1st string, must be in pure unison with the harmonic of the E, 6th string at the fifth fret.
Tuning the 4th string: Play a harmonic on the (in tune) 6th string at twelve, and as this harmonic sounds, adjust the 4th string until the tone E on the second fret is in pure unison.
Tuning the 3rd string: As it is easier to adjust a string while listening to a continuous reference tone, you may first try the following: Play a harmonic on the (in tune) 4th string at twelve and as this sounds, adjust the 3rd string until D at the 7th fret is in pure unison.

Guitar Lessons: Blues: Slide Guitar: Open D Tuning
Open tunings allow the player who often plays alone have a full sound of rhythm and melody underneath his voice since there are no other players to fill these roles.
With an open tuning, you are usually tuning your guitar to a chord (alternate tunings are not tuned to a chord, but rather in a way to emphasize certain notes or drones).
Instead of tuning up to open E (which puts extra strain on the guitars neck), alot of players tune to open D and put a capo on the 2nd fret for open E tuning.

The 6th (low) string or E is tuned down until it is a D, an octave lower than the 4th or D string.
If you're playing open or low and throwing in accents and fills, use the open string and third fret a lot, making that 3rd fret work for you and THEN resolving (or ending) on an open string, probably the G or 3rd string or, better yet, the G or 5th string open.
Once again, the beauty of open tuning is that when you do place the slide across the strings you have, in essence, a major chord to play off of.

 Open D
This tuning is called Open D because when the strings are strummed a D major chord is produced.
Open D is also a good choice for the novice guitarist, since a different complete major chord can be produced simply by barring each fret all the way up the neck.
In addition, because most of the strings are tuned lower than standard tuning, Open D is easier on your instrument's neck than Open E tuning, which in terms of relative tonality is the same as Open D, except each string is tuned one step higher.

Now, when you hear terms like the I, IV, or V and aren't sure what they mean, here's the answer: common chord pattern (one upon which many songs are based) in any key includes chords built on the first (I), fourth (IV), and fifth (V) tones of the major scale.
Here are a few chord suggestions that might work as a substitute, but keep something in mind -- most really fine slide players in open tuning don't even PLAY the V chord -- or, for that matter, often skip the full chord on the IV.
