The arcuate nucleus is the main sensor of circulating levels of leptin, and by projecting to several different regions in the brain, provide the mechanism whereby leptin is capable of integrating a host of responses involved in energy homeostasis.
Signals are then relayed to autonomic regulatory neurons in the parvocellular subdivision of the paraventricular nucleus which through projections to the brainstem or directly to the spinal cord (310), contribute to autonomic mechanisms involved in the generation of fever.
Vagal afferent signals are then carried to the nucleus tractus solitarius in the brainstem, and after projecting to noradrenergic neurons in the ventrolateral medulla (A2), ascend in the ventral noradrenergic pathway and medial forebrain bundle to terminate in the preoptic region.

On the hypothesis that suprachiasmatic neurons exert this control via their projections to paraventricular nucleus, the objective is to use an in vitro brain slice preparation of rat hypothalamus and patch clamp recording techniques to define the characteristics of neurotransmission between the suprachiasmatic nucleus and neurons in the paraventricular nucleus.
The existence of ‘direct’ connections from the suprachiasmatic nucleus to the various subtypes of neurons in the paraventricular nucleus has been at best controversial mainly because the most recent anatomical tracer studies have not been done at the level of resolution (electron microscopic) that would prove their existence.
Therefore, evidence is accumulating to support the hypothesis that suprachiasmatic nucleus neurons do have direct connections to hypothalamic paraventricular neurons associated with cardiovascular functions, suggesting a ‘direct’ mechanism for the biological clock to entrain these functions to a circadian rhythmicity.
