Botany online: Membranes - Compartments - Significance - Peroxysomes - Glyoxysomes
Peroxysomes are usually found in the vicinity of chloroplasts and mitochondria, glyoxysomes occur only near mitochondria.
Especially during growth are a lot of carbohydrates required for the synthesis of new cell walls.
The activities in peroxysomes and glyoxysomes show how important compartmentalization is for the regulation of metabolic pathways.

 Pipecolic acidemia
Pipecolicacidemia formerly described patients with accumulated pipecolic acid in the blood and/or urine.
Pipecolic acid is an intermediate compound in the lysine metabolism that is synthethised by the pipecolic oxidase in peroxysomes.
Recent findings show that the vast majority of pipecolicacidemias are actually due to general peroxysome storage disorders.

In the rat, the peroxysome proliferator activated receptor (PPAR) inducer clofibrate can moderately influence the hormone (testosterone) level and, after single perinatal treatment, irreversibly affects sexual behavior through the mechanism of hormonal imprinting.
The thymic glucocorticoid and estrogen receptors weren't significantly influenced.
The experiments call the attention to the universality of false imprinting by molecules able to bind to the steroid/thyroid receptor superfamily, and point to the different sensitivity to different ligands.

These peroxysomes consume almost half the oxygen of a cell converting it to H
To protect the rest of the cells from this aggressive chemical it is conveniently contained within these membrane bounded spherical vesicles.
Peroxysomes are both involved in the synthesis and breakdown of various classes of lipids and bile acids.

SAV, an archaebacterial gene with extensive homology to a family of highly conserved eukaryotic ATPases.
The new archaebacterial ORF, called SAV (S. acidocaldarius VCP-like) exhibited the usual signature of all members of the family, a highly conserved domain of about 200 amino acids, which is duplicated.
Thus, apart from the VCP-like proteins, SAV also appeared similar, although less clearly, to other ATPases, members of the family, involved in vesicle-mediated transport (NSF, Sec18p), peroxysome assembly (PAS1p), and gene expression in yeast (SUG1p) and in human immunodeficiency virus (TBP-1).
Finally, the discovery of the archaebacterial gene could enlighten not only the evolutionary relationships between the members of this complex ATPase family, but also the cellular function of these proteins, that is presently obscure.

RedOrbit - Health - Calorie Restriction Increases Life Span: A Molecular Mechanism
In mammals, Sirt1, an ortholog of Sir2, controls the metabolism of white adipose tissue.
Lowering of adiposity appears to be one mechanism whereby calorie restriction affects life span.
Seventy years ago, in a remarkable series of experiments lasting nearly 4 years, McCay et al.1 determined the effect of retarded growth of rats on the length of life span and on the ultimate body size.

i381 - Peroxysome proliferator-activated receptor gamma agonist 15-deoxy-delta 12,14 prostaglandin J2 suppresses murine model of experimental autoimmune uveitis
Peroxysome proliferator-activated receptor (PPAR) gamma agonist 15-deoxy-delta 12,14 prostaglandin J2 (15PGJ2) suppresses immune responses in previous studies.
Our data, together with other immunosuppressive properties of 15PGJ2 on RPE cells, suggest that 15PGJ 2 may be a useful candidate for control of intraocular inflammation.

Recently, lipoprotein-lowering agents have been found to be effective for PBC.
The aim of this study was to examine the safety and efficacy of fenofibrate, a member of the fibrate class of hypolipidemic and anti-inflammatory agent via peroxysome proliferatory-activated receptor alpha, in patients with aPBC.
METHODS: Fenofibrate was administered for twelve weeks in nine patients with aPBC who failed to respond to UDCA.

This could be related to the very low basal activity of elongase and desaturase enzymes in endothelial cells, thus preventing arachidonic acid formation from LA in these cells (17).
They include disruption of endothelial cell integrity with alterations of functions of gap-junctions proteins (18), cell morphology (19), membrane-bound enzymes (20), peroxysome (21), and induction of apoptotic cell death (22).
Last but not least, accumulating evidences suggest that increase in intracellular oxidative stress and calcium levels (10,23) could be the main mechanisms by which LA exerts its harmful effects on endothelial cells.
