It is suggested that the multiple enzyme inhibition caused by administration of high doses of phenelzine accounts for its unusual effects on striatal p-tyramine levels compared with other MAO inhibitors, i.e., its initial lack of effect on p-tyramine levels followed later by very large increases in p-tyramine levels.

Short-term phenelzine therapy may not wipe out social fears entirely, adds Liebowitz, but it allows patients to talk to other people, develop skills and build close relationships, often for the first time.

We started the trial when Dr. Joseph Chu, who was then a medical student, realized that CFS like symptoms followed treatment of high blood pressure with a pill that altered firing of a part of the brain known as the locus coeruleus.
Since the antidepressant phenelzine returned locus coeruleus firing to normal, Dr. Chu reasoned that phenelzine might be helpful in the treatment of CFS.
Since only low doses of phenelzine are required to alter locus firing, we elected to do the treatment trial with doses that would probably have no antidepressant effect.
