Photochemistry studies of phenyl substituted-1,2,4-thiadiazoles have revealed that 5-phenyl-1,2,4-thiadiazoles 31, 90, 98, 54 and 47 undergo a variety of photochemical reactions including photofragmentation, phototransposition, and photo-ring expansion while irradiation of 3-phenyl-1,2,4-thiadiazoles 46, 105 and 106 leads mainly to the formation of photofragmentation products.
The photoreactivity of these phenylthiadiazoles in acetonitrile is substantially decreased when the phenyl ring at position 4 is substituted with an electron donating or withdrawing group.
The relaxed S1 would be the state from which the observed photoproducts originate and the observed fluorescence with the smaller Stokes’ shifts compared with the Stokes’ shifts observed in acetonitrile.

Conversely, if the N atom carries an alkyl substituent while a phenyl ring is attached to a C-atom of the aziridine, oxidation results in spontaneous ring opening to yield azomethine ylide radical cations which have broad absorptions in the 500-800 nm range.
The results of quantum chemical calculations confirm the assignment of these species and allow to rationalize the different effects that phenyl rings have if they are attached in different positions of aziridines.

Their activity on rat-insulin-secreting cells and rat aorta rings was compared to that of the K(ATP) channel activators (+/-)-cromakalim, diazoxide, (+/-)-pinacidil, and compound 4.
Structure-activity relationships indicated that the most pronounced inhibitory activity on the pancreatic tissue was obtained by introducing a meta- or para-electron-withdrawing group (a chlorine atom) on the C-4 phenyl ring (drugs 37-42).
