The phospholipase C system which generates diacylglycerol and IP3 from phosphatidylinositol 4,5-bisphosphate has been extensively studied in recent years.
These include the protein kinase C regulator diacylglycerol, the calcium signal inositol 1,4,5-trisphosphate, probably phosphatidic acid, and phosphorylated forms of phosphatidylinositol.
Diacylglycerol generation was also activated by a protein kinase C activator, PMA.

The level of phosphatidylinositol-4, 5-bisphosphate in spleen lymphocytes was unaffected by diet.
The fish or olive oil diets, and to a lesser extent the safflower or evening primrose oil diets, decreased the adhesion of both freshly prepared and Con A-stimulated lymphocytes to macrophage monolayers.
Morphological analyses showed that evening primrose oil diminished endothelial lesions caused by the atherogenic diet, reducing area of the arterial wall (from 6.9 +/- 0.2 to 4.7 +/- 0.2 microm2 x 10(6)) and the degree of neointimal proliferation (from 0.6 +/- 0.02 to 0.4 +/- 0.09 microm2 x 10(6)).

EPR measurements using a spin-labeled derivative of PI(4,5)P^sub 2^ indicate that the MARCKS-ED binds ~3 molecules of PI(4,5)P^sub 2^, and site-directed spin labeling of the MARCKS-ED peptide suggests that neither specific van der Waals contacts nor hydrogen bonds are required for the pcptide-PI(4,5)P^sub 2^ interaction (Rauch et al., 2002).
SDSL is used to position the peptide in the membrane interface and suggests that the interactions with PI(4,5)P^sub 2^ are electrostatic in origin.
Site-directed spin labeling of the SCAMP-E peptide indicates that the position and structure of membrane bound SCAMP-E are not altered by the presence of PI(4,5)P^sub 2^, and that the peptide backbone is positioned within the lipid interface below the level of the lipid phosphates.
