  Phytohormone

Title of Invention: Pentasaccharide phytohormones a nd methods for their use
A phytohormone for inducing root hair curling and root nodulation in the roots of leguminous plants particularly in the absence of nitrogen-fixing bacteria.
Biological activity of the phytohormone were scored on a scale of 1 to 5 with 5 exhibiting marked deformations and swelling of the root hairs.
Phytohormone activity was detectable, a grade of 3 or higher on the scale, down to a concentration of at least 10.sup.-12 M. At concentrations of greater than about 10.sup.-3 M, the phytohormone exhibited an inhibitory effect with respect to root hair curling.

Summary: It is shown that the phytohormonal factor of serum, as measured by its inhibitory effect on the growth of plant root cells, is in part free and active and in part bound in an inactive state to serum proteins.
The lupine root method is described, and it is demonstrated through a number of systematic investigations that during the preliminary germination of the roots a number of factors influence the rate of growth of the roots.
Testing of the phytohormonal effect of urine and denatured serum by the coleoptile method characterizes the phytohormones in these liquids as being essentially of the auxin type, and spectrophotometric examinations show correlation between phytohormonal effect and quantity of indole compounds in urine.

 Phytohormone

This study investigated the physical manifestations of the responses of Arabidopsis thaliana wild type and phytohormonal mutants subjected to salt stress.
The phytohormonal mutants that were studied included plants with altered ethylene, abscisic acid, auxin, and brassinosteroid biosynthesis or perception.
However, based on the results of this study it can be concluded that in order to produce salt stress tolerant plants one should use mutations not in one phytohormone, but perhaps in few: The results of this experiment suggested that one should use a combination of ABA resistance, ethylene insensitive and BR overproducer mutations.

By quantifying the parameters cell-elongation growth and cell-division frequency under various phytohormone regimes in transgenic tobacco BY-2 cell suspensions, we showed that the relative rates of both cellulair processes become coupled by overexpression of ENOD40.
Only a small number of phytohormones are sufficient to regulate virtually every aspect of plant growth and development.
Our data support understanding of how so few phytohormones can regulate such a diversity of processes, and which other components are involved in the regulatory mechanisms.

Relatively non-volatile acidic phytohormones such as salicylic acid, jasmonic acid, indole-3-acetic acid, abscisic acid are converted into volatile methyl esters and easily separated from the extraction mixture by collecting the analytes as volatiles.
This facile and rapid method for analyzing a wide range of phytohormones, elicitors, and chemical signals will enable elucidation of key crosstalk interactions and identify molecular targets for increasing the inducible expression of plant defenses.
A bottleneck in crosstalk research is the quantification of numerous interacting phytohormones and regulators.

Investigations aimed at understanding the mechanisms that determine the "strength" of phytohormone signals, including processes that regulate the levels of phytohormones by means of biosynthesis, metabolic activation and inactivation, transport across cell membranes and cellular localisation.
Analysis the functions of the actin and tubulin cytoskeleton, and the role of regulatory proteins in phytohormone transport and action.
The auxin-, abscisic-acid- and cytokinin-types of phytohormones bind to columns of the MCX sorbent and can be eluted stepwise using solvents supplemented with various concentrations of methanol and ammonium hydroxide.

CYTOKININS, ABSCISIC-ACID AND LIGHT AFFECT ACCUMULATION OF CHLOROPLAST PROTEINS IN LUPINUS-LUTEUS COTYLEDONS WITHOUT ...
The effects of cytokinin, abscisic acid, gibberellic acid (GA(3)) and indolylacetic acid (IAA) have been studied at the ultrastructural, steady-state mRNA and protein levels using 15 gene-specific probes for plastid proteins and corresponding antisera.
These results indicate a protein-specific response to phytohormones, which can differ even for polypeptides belonging to the same membrane complex.
They also suggest different modes of interaction between hormones and light, quite different phytohormone action in the two compartments, and demonstrate that phytohormones influence the biogenesis of the thylakoid membrane mainly posttranscriptionally.
