JEFFREY  PATTON, Ph.D.   (Site not responding. Last check: 2007-10-20)
All of the current projects in Dr. Patton's laboratory are concerned with the splicing of premessenger RNA (premRNA).
The metabolism of cofactors that are necessary for premRNA splicing is one of the current interests of Dr. Patton's laboratory.
In addition, the gene expression by the alternative premRNA splicing of HPV transcript is another focus of the laboratory.

JBPC Journalclub » Spliceosomal introns in the deep-branching eukaryote Trichomonas vaginalis.   (Site not responding. Last check: 2007-10-20)
Eukaryotes have evolved elaborate splicing mechanisms to remove introns that would otherwise destroy the protein-coding capacity of genes.
Nuclear premRNA splicing requires sequence motifs in the intron and is mediated by a ribonucleoprotein complex, the spliceosome.
Here we demonstrate the presence of a splicing apparatus in the protist Trichomonas vaginalis and show that RNA motifs found in yeast and metazoan introns are required for splicing.

 Monaco 2004
As the dystrophin gene itself will not be altered, the effect of the antisense treatment will not be permanent, so it will have to be repeated at intervals still to be determined.
For this new method, the U7-snRNAs were genetically modified so that they no longer bound to the histone premRNA but only to the splice sites in the region of exon 23 of the pre-mRNA of the mouse dystrophin.
To achieve this, a gene coding for the modified U7-snRNA together with a creatine kinase enhancer was packed into plasmids as vectors and then transferred into isolated myoblasts of the mouse.

Anchor binding domain: A 33kDa editosome protein that has RNA recognition and glycine-rich motifs and a 44kDa protein essential for editing form the main core of this domain, these proteins help in the stability of the complex. The anchor duplex is bound by this domain of the model.
Upstream stablization domain: This doamin helps in the stable interaction between the upstream region of premRNA and 3'region/Utail of gRNA.
Catalysis domain: After the orintation of premRNA to be edited and gRNA that specify the editing to editosome, catalysis domain which conatins the editing enzymes catalyses the insertion or the deletion as directed by gRNAs.

Effects of feeding on hepatic GRP78 mRNA and premRNA abundance in mice.
Hepatic RNA was hybridized either to probe 1, which produced a 257-base protected fragment (labeled GRP78 premRNA in lanes 7–12), or to probe 2, which produced a 200-nucleotide protected fragment (labeled GRP78 premRNA in lanes 13–18).

Splicing 2003 will highlight the potential impact of alternative RNA splicing on drug discovery
Alternative RNA splicing is a tightly regulated process that controls overall protein functionality and diversity.
Notably, research will be presented regarding the Drosophila Down Syndrome cell adhesion molecule gene (Dscam), where a single premRNA can generate more than 38,000 different transcripts, highlighting the complexity of the regulatory system that governs the alternative RNA splicing process.
Dr Bruno Tocqué, CEO of ExonHit Therapeutics, commented: “I am very pleased that we have been able to assemble this distinguished group of scientists to present at Splicing 2003.
