  Primer
A primer is required because most DNA polymerases (enzymes that catalyze the replication of DNA) cannot begin synthesizing a new DNA strand from scratch, but can only add to an existing strand of nucleotides.
This RNA is produced by an RNA polymerase, and is later removed and replaced with DNA by a DNA polymerase.
Primers that are too short would anneal at several positions on a long DNA template, which would result in non-specific copies.

All of these primers were identified and tested by our own lab based on consensus between the published large and small subunit RNA sequences from fungi, plants and other eukaryotes; sources of other useful primer sequences from published literature are also indicated.
The nuclear-encoded ribosomal RNA genes (rDNA) of fungi exist as a multiple-copy gene family comprised of highly similar DNA sequences (typically from 8-12 kb each) arranged in a head-to-toe manner.
Depending on the orientation and position of the 5S RNA gene, the PCR may be used to sequentially amplify either aportion of the intergenic spacer region (IGS) beyond the large subunit RNA coding region.

When either the homologous RNA (avian myeloblastosis virus RNA) or a heterologous RNA (poliovirus RNA) was used as a template, the anticomplementary DNA synthesized in vitro by avian myeloblastosis virus reverse transcriptase (RNA-directed DNA nucleotidyltransferase, EC 2.7.7.7) was primed by fragments of the original RNA template that usually had adenosine at their 3' ends.
When we used phage T4 RNA ligase (EC 6.5.1.3) to label the 3' end of the RNA template fragments contained in the RNA· cDNA hybrid intermediate, adenosine was found to be the principal nucleoside carrying the label.

RNA Polymerase does not need a 3'OH to produce RNA or RNA primers de novo.
