RasGAP is a Caspase 3 substrate (1) and acts as both pro- and anti-apoptotic protein depending on the extent of its cleavage by caspases (2).
At low levels of caspase activity, RasGAP is cleaved at position 455, generating an N-terminal fragment (fragment N) and a C-terminal fragment (fragment C).
Fragment N appears to be a general blocker of apoptosis downstream of caspase activation because it inhibits caspase 9-induced cell death.

RasGAP is a regulator of Ras- and Rho-dependent pathways but also a caspase-3 substrate that bears two cleavage sites that are used sequentially as caspase activity increases.
The survival of cells having mildly activated caspase-3, in response to low stresses for example is critically dependent on the partial cleavage of RasGAP into fragment N. The second cleavage of RasGAP results in the destruction of fragment N and its anti-apoptotic activity.
RasGAP is the first example of a caspase substrate that, when cleaved, negatively or positively regulates apoptosis in a manner that is dependent on the extent of caspase activation.

With the use of a Raf-1-kinase pull-down assay, the defective MAP-kinase response in TSAd-deficient T cells was attributed to impaired switching of the small G-protein, Ras, from an inactive GDP-bound to an active GTP-bound form.
Consistent with this, RasGAP activity remained at constitutively high levels in TSAd-deficient T cells after TCR stimulation although was turned-off in control T cells in response to the same stimulus.
Thus, TSAd is identified as a novel inhibitor of RasGAP that is necessary for activation of the Ras-MAP-kinase pathway in T cells.

Using specific antibodies, we found that phospholipase C-gamma 1 and the p21ras GTPase-activating protein, rasGAP, are present in human platelets.
Our results suggest the formation of a signaling complex involving rasGAP, phospholipase C-gamma 1, and rap1B that might be important in the cascade leading to platelet activation.
Although thrombin stimulation had no effect on the level of phosphorylation of phospholipase C-gamma 1 and only slightly increased the tyrosine phosphorylation of rasGAP, the agonist induced the association of rasGAP with rap1B, as indicated by the appearance of rap1B on a Western blot of rasGAP immunoprecipitates.

Recent studies form our and others laboratories have recently identified a major role for Cdk7 and RNA polymerase II incell survival, thus, potentially linking the hypertrophy pathway to that of cell survival.
Current work in the laboratory aims to further delineate the role of RasGAP in cell survival and its relationship to hypertrophy both in cultured cardiac myocytes and in transgenic and knockout mouse models.
