Restriction enzymes are isolated from a wide variety of bacterial genera and are thought to be part of the cell's defenses against invading bacterial viruses.
Isolation of sufficient DNA for RFLP analysis is time-consuming and labor intensive.

Restriction fragment length polymorphism (or RFLP) is a property of DNA used in molecular biology methods, for example, genetic fingerprinting or paternity testing.
The restriction fragments are then separated according to length by agarose gel electrophoresis.
The distance between the locations cut by restriction enzymes (the restriction sites) varies between individuals: so the length of the fragments varies, and the position of certain gel bands differs between individuals (thus polymorphism).

Restriction fragment length polymorphisms (RFLPs) is the term scientists use to denote that, when DNA is cut by restriction enzymes fragments of DNA are produced.
The RFLP technique is considered more accurate than the PCR method, because it requires variations of the gene sequence that are found on longer strands.
In other words, RFLP cannot be used in situations where very small strands of DNA are present, such as a crime scene, where a blood stain or a small piece of hair are the only evidence discovered.
