Grenville was a cousin of both Sir Walter Raleigh and Sir Francis Drake, and grandfather of Sir Richard Grenville, of English Civil War notoriety.
Grenville had planned to circumnavigate the globe in the late 1570s, but Queen Elizabeth gave the honour to Sir Francis Drake instead.
The last and fatal fight of Grenville and the Revenge is commemorated in a poem by Alfred Tennyson ("The Revenge") and a song by Al Stewart ("Lord Grenville").

At an early age Grenville is supposed to have served in Hungary under the emperor Maximilian against the Turks.
Grenville himself soon returned with the fleet to England, capturing a Spanish vessel on his way, but in 1586 he carried provisions to Roanoke, and finding the colony deserted, left a few men to maintain possession.
But, either from some misuiiderstanding of the order, or from some idea of Grenvilles that the Spanish vessels rapidly approaching were the ships for which they had been waiting, the Revenge was delayed and cut off from her consorts by the Spaniards.

However, Grenville did not have to deal with the colonial problems that resulted in his policies because he was invited to resign by George III in July 1765.
All these factors led to the fall of Grenville's ministry in July 1765 when he was persuaded to resign, to make way for a ministry headed by the Duke of Cumberland with the Marquis of Rockingham as First Lord of the Treasury.

Grenville returned to Virginia in 1586, with stores for the colonists, but they had left prior to his arrival.
In 1591 Grenville was appointed Vice-Admiral and second-in-command of a naval force under Lord Thomas Howard, which was sent to the Azores to await for the homeward bound Spanish treasure fleet.

Next, Grenville – who had been knighted by Elizabeth in about 1577 – was fired by Sir Walter Ralegh's plans for a colony in North America, and in 1585, he sailed to Virginia with 300 settlers on board.

In 1591 Grenville sailed to the Azores, with Lord Thomas Howard, as second-in-command of an expedition sent to capture a Spanish treasure fleet.
His ship, the Revenge, was cut off from the rest of the English fleet and was captured by the Spanish after a battle in which Grenville was mortally wounded.
