In 1501, accompanied by Balboa and Juan de la Cosa, he discovered the mouths of the Magdalena River.
Because of difficulties with the Spanish crown, it was 1525 before he returned to found Santa Marta.

Bastidas was familiar with the New World, since he had accompanied the Admiral, Don Cristóbal Colón on his second voyage, and also with Alonso de Ojeda, on his first voyage.
Bastidas was charged with illegally trading with the natives without a license, and his original commision only allowed him to trade in the lands that he would discover.
Bastidas and de la Cosa, were each awarded and annual pension of 50,000 maravedis for their discoveries, and was given the title of Governor, properties, and royalties for the land he discovered.

The partners in this enterprise were Rodrigo de Bastidas (a Seville merchant) and Juan de la Cosa, formerly a pilot with Columbus.
During their first voyage from the Guajira to the Isthmus of Panama, Bastidas and de la Cosa discovered and named the harbour of Cartagena (later to become one of Spain's principal Caribbean ports), traded with the chiefs of the Sinú region, and returned with a good quantity of gold from the Gulf of Urabá.
As early as 1529, an expedition under Pedro de Lerma was offering the Taironas agricultural tools of iron, and also 'many beads, many combs, knives and scissors, coloured hats, caps, and shirts finely worked at the neck'.

In 1501 the Spaniard Rodrigo de Bastidas, in the company of Juan de la Cosa and Vasco Núñez de Balboa, was the first European to explore the Atlantic coast of the Isthmus of Panama.
In 1510 Diego de Nicuesa, another Spanish explorer, established the settlement of Nombre de Dios at the mouth of the Chagres River, and to the southwest, Alonso de Ojeda founded San Sebastian de Urabá.
With the final destruction of Nombre de Dios in the late 16th century by the Englishman Francis Drake, commercial activity was moved to the hamlet of Portobelo, overlooking the calm bay recorded by Christopher Columbus in 1502.

Vasco Núñez de Balboa, a member of Bastidas's crew, had settled in Hispaniola (present-day Dominican Republic and Haiti) but stowed away on a voyage to Panama in 1510 to escape his creditors.

Vasco Nunez de Balboa was a poor uneducated man born in 1475 in Jerez de los Caballeros in Estremadura.
In 1501 he set sail on an expedition commanded by Rodrigo de Bastidas to explore the northern coast of present-day Colombia.
This new colony, Santa Maria de la Antigua, was founded by Nicuesa in present-day Panama during the year 1510.
