Selegiline (MAO inhibitor)   (Site not responding. Last check: 2007-11-04)
Selegiline's duration of action depends on the time required to regenerate MAO type B. At higher selegiline doses (e.g., 20÷40 mg/day), brain MAO is blocked nonselectively, predisposing patients to the risks of traditional MAOIs that block MOA type A (e.g., hypertension).
: Selegiline is administered orally and is readily absorbed from the GI tract and crosses the blood/brain barrier.
Selegiline is eliminated slowly by the kidneys; about 45% of a single 10 mg dose is eliminated in the urine as the three active metabolites in 48 hours.

 Selegiline
Selegiline, because it protects the section of the brain that produces dopamine, may have a protective effect on the decay of the brain brought on by dopamine deficiency.
Selegiline is therefore free of restricting "cheese effects," yet is able to increase brain dopamine by preventing its normal breakdown within nerve cells and in the gaps between nerve fibers.
Increase in depression in the elderly, age-dependent decline in male sexual vigor and the frequent appearance of Parkinsonian symptoms in the latter decades of life may be due to a decrease of dopamine and trace amines in the brain.

Selegiline increases the levels of dopamine in the striatum (a brain area dependent on the dopamine signal) of rats [2].
Selegiline can be used as monotherapy and as a levodopa adjunct in patients with PD and increases the levels of dopamine in their striatum [4].

Selegiline for Parkinson's disease - [Medication]
Selegiline prolongs the effects of dopamine in the brain by preventing its breakdown.
Selegiline may be used in the early stages of Parkinson's disease to treat very mild symptoms (such as resting tremor) and delay the need for levodopa.
In people with advanced Parkinson's disease who are taking levodopa, selegiline may be added to levodopa treatment to reduce motor fluctuations, increase the time of effect of the levodopa, and decrease the amount of levodopa needed to control symptoms.

Research Page: Effects of Selegiline on craving for nicotine
Selegiline is a medication which helps to increase the amount of this chemical in the brain.
We will determine whether selegiline is useful by measuring the extent to which the medication reduces nicotine craving after study participants stop smoking and the extent to which participants are able to remain off cigarettes for the duration of the study..
Selegiline is of interest because this medication may be able to reduce cravings that increase the likelihood that smokers will relapse after quitting.

Selegiline can indirectly raise norepinephrine because dopamine can be catabolized within the brain to norepinephrine, although the extent of this is variable.
Selegiline is partly metabolized to an inactive stereoisomer of methamphetamine in vivo in levels that, even if active, most likely are far too low to have any significant effect.

Selegiline (Eldepryl), a selective monoamine oxidase (MAO) inhibitor, has recently been approved by the Food and Drug Administration as an adjunct to carbidopa/levodopa (Sinemet) therapy in the management of Parkinson's disease.
Furthermore, selegiline is transformed to amphetamine and methamphetamine, an action that also interferes with neuronal reuptake and causes release of biogenic amines such as dopamine.
The exact role of selegiline in the therapy of Parkinson's disease remain,, to be determined.

Addition of selegiline to levodopa therapy may enable the dosage of levodopa to be reduced by an average of 30%.
Although some critics have suggested that selegiline is simply providing symptomatic relief in disease which continues to progress (9-11) the promising preliminary results have been suggested by others to justify the prescription of selegiline 10 mg daily to patients, especially younger ones, in the early stages of Parkinson's disease, in the hope of impeding progression.

 Selegiline hydrochloride
Selegiline is usually used in combination with a medicine called levodopa to treat Parkinson's disease.
Selegiline can be used to prolong the effect of levodopa, because it stops the dopamine that is formed from the levodopa from being broken down in the brain.
Selegiline should not be started until at least two weeks after stopping these antidepressants and until at least five weeks after stopping fluoxetine.

Abstract from Selegiline for Smoking Cessation pilot project -News - CENTURY
Background: Since dopaminergic mechanisms appear to be involved in nicotine dependence, we studied the safety and efficacy of the monoamine oxidase B inhibitor selegiline hydrochloride compared with placebo for smoking cessation in nicotine-dependent cigarette smokers.
As part of a Yale University study, 20 smokers were given selegiline hydrochloride (L-Deprenyl or Eldepryl) and 20 were given a placebo; both groups attended weekly counseling.
Selegiline inhibits the enzyme monoamine oxidase B (MAO-B), that normally breaks down dopamine, keeping dopamine levels from falling.

 Selegiline in Parkinson's Disease
Selegiline does delay the need for levodopa therapy, however, the onset of disability or levodopa-induced motor fluctuations is not postponed by early selegiline treatment.
While most observers now accept that selegiline does not have a neuroprotective effect, there is still a body of animal studies supporting this possibility, and some clinicians have continued to argue that selegiline may retard the progression of Parkinson’s disease.
Selegiline may be of use for patients with motor fluctuations although it is not necessarily better than alternative strategies such as slow-release levodopa, dopamine agonists or COMT inhibition.

Selepryl (Selegiline), is a drug used as an adjunct therapy of Parkinson's disease due to it's anti-neurodegenerative properties it is biochemical defined as a selective monoamine oxidase type B (MAO B) inhibitor, Selegiline was originally developed as an antidepressant agent.
Selegiline turned out to be an efficient inhibitor of the uptake of dopamine in the striatum and this effect was found to play an important role in the facilitation of the activity of the nigrostriatal dopaminergic neurons in animals treated continuously with (-)Selegiline.
Selegiline in striking contrast with the MAO inhibitors used previously inhibits the noradrenaline releasing effect of indirectly-acting amine in vascular smooth muscle.

A continuation of the study, in which140 patients received selegiline or placebo in combination with individually tailored levodopa therapy, found that selegiline, compared with placebo, significantly slowed the progression of disease disability as measured by the Unified Parkinson Disease Rating Scale (UPDRS) total score or by motor and Activities of Daily Living subscores.

Selegiline, therefore, increases the amount of dopamine in the striatum and optimizes the ability of the remaining dopamine to cause the signal for movement in patients with PD.
When selegiline is given to patients with PD who have not yet received any medication for PD, it is effective and can delay the need to take levodopa.
The long-term use of selegiline therapy may prolong the clinical benefits of levodopa; selegiline therapy can last for up to 36 hours and so it can sometimes reduce the fluctuations in effectiveness of levodopa that people may experience after the first few years.

But those patients who were taking selegiline in combination with levodopa were no more likely to die during the study than the people without Parkinson's.
Selegiline has been used as a treatment for Parkinson's for nearly 25 years, but excitement about the drug peaked in the mid-1980s when research suggested that it may have neuroprotective effects, thereby slowing the effects of the disease on the brain.
