The semicircular canals are three half-circular, interconnected tubes located inside each ear that are the equivalent of three gyroscopes located in three planes perpendicular (at right angles) to each other.
Each canal is filled with a fluid called (The bodily fluid that fills the membranous labyrinth of the inner ear) endolymph and contains a motion sensor with little hairs whose ends are embedded in a gelatinous structure called the cupula.
The cupula and the hairs move as the fluid moves inside the canal in response to an angular acceleration.

Netrin 1 is required for semicircular canal formation in the mouse inner ear.The morphogenetic development of the mammalian inner ear is a complex multistep process, the molecular and cellular details of which are only beginning to be unraveled.
In netrin 1 mutant mice, fusion plate formation is severely affected resulting in a reduced anterior semicircular canal and the complete lack of the posterior and lateral canals.
Our results suggest that netrin 1 facilitates semicircular canal formation through two different mechanisms: (1) it participates in the detachment of the fusion plate epithelia from the basement membrane, and (2) it stimulates proliferation of the periotic mesenchymal cells which then push the epithelial cell walls together to form the fusion plate.

Ewald introduced a surgical procedure in the late 1800s termed "canal plugging" where the slender duct of a semicircular canal is occluded in an attempt to block sensitivity of the associated ampullary organ to angular motion stimuli (Ewald, 1892).
Since its reintroduction by Money and Scott (1962) canal plugging has been used for the treatment of vestibular disorders such as benign paroxysmal positional vertigo and to study the influence of individual canal and otolithic afferent inputs to the vestibular reflex systems.
Until recently it was presumed that canal plugging completely blocked the sensitivity of the occluded canals and that partial recovery of the VOR following surgery was the result of adaptation by the central nervous system to use inputs from the remaining functional end organs.

To test if the nystagmus results from an imbalance of the central pathways of the semicircular canals we examined four patients with dorsolateral medullary syndrome (two right, two left sided) and recorded nystagmus binocularly with the three-dimensional search coil technique.
