Alex Poole - Literature Review - Serif vs. Sans Serif Legibility
Serifs are used to guide the horizontal "flow" of the eyes; The lack of serifs is said to contribute to a vertical stress in sans serifs, which is supposed to compete with the horizontal flow of reading (De Lange et al., 1993)
What ever their origin, serifs have been around for so long that perceived legibility is very likely to have been affected by familiarity - readers tend to rate as more legible the typefaces they are most used to (Tinker, 1963 ; Zachrisson, 1965).
It is of course possible that serifs or the lack of them have an effect on legibility, but it is very likely that they are so peripheral to the reading process that this effect is not even worth measuring (Lund, 1999).

It is probably there from that the serif was born—the cut letter kept the form of the letter that had been set out by the brush.
Another reason for the serif’s preservation in the stone-cutting process was the type of marble that the Romans were using in the first place.
The first typeface serifs were softly modulated—a smooth curve connected the stem to the serif.

Serifs are the small strokes added to the main stokes of a letterform.

Serif vs. Sans-Serif Fonts: In Search of the Perfect Font
In print, serifs are thought to enhance readability and legibility because they help readers discriminate the ends of letters and make the form of each letter more complex (Arditi, Cho 2005).
In a recent study of serif legibility and readability, Arditi and Cho evaluated the use of serifs with respect to reading speed, letter recognition, and continuous reading on paper.
In the end, the researchers found that one of the perceived advantages of serif fonts, which is that serifs act as letter distinguishers, may be irrelevant on the web.

The World of Fonts. Sans Serif History - webreference.com
It is interesting to note that the development of sans serif typefaces in this century went in a direction opposite to that of the serif type development of previous centuries.
Conversely, sans serif fonts started from Futura with its artificial look, then were for a long time dominated by neutral "transitional" Helvetica, and recently a number of distinctively liberal (and, in some classifications, even termed "humanist") sans serif faces became popular.
Thus, the 20th century process of sans serif humanization is a negation, a mirror image, a contrasting parallel for the earlier process of serif dehumanization in 15th-19th centuries---just as sans serif itself is a contrasting match for serif.

This experiment found that the serif font Times New Roman was preferred overall and in terms of personal usability and web usability to the ornate sans serif font.

 TYPOGRAPHY: Typefaces
A serif typeface like Times New Roman (the default text face in most Web browsers) is about average in legibility on the computer screen, with a moderate x-height.
The most conventional scheme for using typefaces is to use a serif face such as Times New Roman or Georgia for body text and a sans serif face such as Verdana or Arial as a contrast for headlines.
If you choose to combine serif and sans serif faces, select fonts that are compatible and don't use more than two typefaces (one serif, one sans serif) on a page.
