DNA replication occurs in the nucleus.  The cell copies and transmits genetic information via a process known as “semi-conservative replication.”  Each strand of the helix, or each side of the ladder, serves as a template for a new complementary and anti-parallel molecule as seen in slide 7.
The enzymes, primer and nucleotides are also used in PCR (polymerase chain reaction) and other amplification technologies, so this is an important process to know to better understand the principle of these technologies.
In slide 8 a double stranded DNA molecule has been unzipped or H bonds have been broken by helicase.  The primer needed to begin replication always attaches to the 3’ end of the template strand.

How do you do the Twist/Double Circle Boondoggle stitch?
To start this stitch, you'll need three strands of boondoggle, two of the same length, and a third that is approximately 1½ times as long as one of the first two.
Continuing, grab the next short strand, on the opposite side, and bring it UP and OVER the first, then ACROSS and UNDER the second loop.
That strand was TWISTED and would have caused the finished stitch to have an odd appearance when you compared it to all the other stitches.

Synthesis of new strands occurs at the replication fork, the place where the DNA helix is being separated into two template strands.
For this strand, an complementary RNA primer is synthesized near the replication origin, and DNA polymerase then extends the new strand from its 5' end to its 3' end as one continuous strand.
When a section of template strand is separated, primase synthesizes a short primer, beginning near the junction of separated and unseparated strands.
