Busy brains may stave off Alzheimer's signs
Sisodia says exercise, along with any kind of mental activity from reading to doing the crossword puzzle, are probably the equivalent of chew toys and running wheels for humans.
Sisodia and colleagues were intrigued by recent research suggesting that connections between brain cells change throughout life in response to new environments, and that activity at these communication hubs can affect the production of b-amyloid peptides.
Sisodia believes these genes may hold clues to exactly how the enriched mice were able to lower their b-amyloid peptide levels.

Sisodia - Wikipedia, the free encyclopedia
The Sisodia (also known as Shishodya or Sisodya or Sisodhya) are a Rajput clan who ruled the kingdom of Mewar in Rajasthan.
Hamir Singh (1326-1364) recaptured Chittaurgarh in 1326, and was the first ruler of the clan to use the royal title of "Rana." He changed the family name to Sisodia, derived from Sisoda, the name of the village where he was born.
After the Mughal emperor Aurangzeb attacked the neighboring kingdom of Marwar in 1679, the Sisodias formed a triple alliance with the Rathores of Marwar and the Kachwahas of Jaipur to liberate the Rajputs from the Mughals.

 Sangram Sisodia
Prior to joining The University of Chicago, Dr. Sisodia was a Professor of Pathology and Neuroscience at The Johns Hopkins University School of Medicine where he defined mechanisms by which the amyloid (Aβ) protein precursor, or APP, is transported and processed in mammalian cells and in the nervous system.
Over the years, Dr. Sisodia's group has made important discoveries related to the biology of presenilins, proteins that are mutated in the major of cases of early onset-familial Alzheimer's disease (FAD).
Sisodia received his Ph.D. in Biochemistry from the University of Georgia and joined Johns Hopkins as a Postdoctoral Fellow in 1985 where he rose to the rank of Professor of Pathology and Neuroscience.

Protein Glitch May Be Early Problem In Inherited Alzheimer's
Sisodia and Gopal Thinakaran, Ph.D., David Borchelt, Ph.D., and Michael Lee studied presenilin in cell cultures and in mice that contained copies of the human presenilin gene.
Because presenilin is present only at very low levels, Sisodia notes, it's unlikely to be directly causing the widespread damage to nerve cells seen in FAD, but it may interfere with other proteins that could.
According to Sisodia, mutated forms of presenilin may influence the formation of plaques in the brain that are among the earliest signs of Alzheimer's disease.

Sisodia believes that over the next five years or so, studies will show that it's the ability to clear amyloid--either before or after it's deposited in brain plaques--that distinguishes the genes of some of those who get AD from those who don't.
Sisodia adds that the mouse models are "spectacular in terms of amyloid deposition, and even for formation of neurofibrillary tangles.

The levels of b-amyloid peptides, which clump together to form the brain "tangles" or plaques that are toxic to nerve cells in Alzheimer's disease, were significantly lower in the enriched mice.
More experiments with larger numbers of animals are needed to determine exactly how enriched environments benefit mice, whether through increased physical activity, a boost in visual, social, and spatial stimuli that awaken the brain, or some combination of all of these factors.
Sisodia and colleagues intrigued by recent research which suggests that connections between brain cells change throughout life in response to new environments, and that activity at these communication hubs can affect the production of b-amyloid peptides.

Speaking at the Bankers' Conference 2004, Sisodia also suggested that the government should improve on its ownership role by demanding a better return on assets, strengthening the functioning of bank boards and making the management more accountable.

University of Chicago Hospitals: Enriched environment delays onset of Alzheimer's in mice
Mice from enriched settings also had more of an enzyme that breaks down amyloid as well as increased activity of several genes involved in learning and memory, brain cell survival and the growth of new blood vessels.
Sisodia, and colleagues from his lab -- Orly Lazarov, PhD, assistant professor of neurobiology, pharmacology and physiology and lead author of the study, and John Robinson, senior research technician -- studied mice carrying two mutated genes (amyloid precursor protein and presenilin-1) that predispose the animals to develop Alzheimer's disease early in life.
A lot of the process involves simple plumbing, Sisodia suspects, delivering blood to the brain and carrying harmful substances away.

Mental and Physical Exercise Benefit Mice with Alzheimer's
Mice who keep their brains and bodies busy in an "enriched" environment of chew toys, running wheels and tunnels have lower levels of the peptides and brain plaques associated with Alzheimer's compared to mice raised in more sparse conditions, according to a new study in the March 11 issue of the journal Cell.
Levels of b-amyloid peptides, which clump together to form the brain "tangles" or plaques that are toxic to nerve cells in Alzheimer's disease, were significantly lower in the enriched mice, according to Sisodia and colleagues.
The enriched mice may have been better equipped than their less-stimulated counterparts to sweep these peptides out of the brain, suggests the researchers' analysis of gene and enzyme expression in the animals.
